1. Chondrocyte-matrix interactions: with emphasis on the collagen binding integrin a10ß1
Sammanfattning : The work in this thesis has been focused on chondrocyte integrins and their interaction with the matrix proteins chondroadherin (CHAD) and collagen type II (CII). Affinity purification of CHAD-binding proteins from chondrocyte lysate, followed by immunoprecipitation of EDTA-eluted material identified alpha2beta1 as a CHAD-binding integrin. LÄS MER
2. Articular cartilage proteoglycan biosynthesis and sulfation
Sammanfattning : Glucosamine (GlcN) and glucosamine sulfate (GS) have been used to treat the patients with osteoarthritis(OA) as a disease-modifying agent. Previousin vitro studies have focused on the effects of GlcN or GSon cartilage metabolism, whereas in vivo studies have investigated their potential for the treatment of OA. LÄS MER
3. The role of autophagy in cartilage physiology and metabolism : implications for growth and ageing
Sammanfattning : Cartilage is the main constituent of the embryonic skeleton. At the ends of long bones cartilage forms a growth plate consisting of chondrocytes in distinct stages of differentiation and arranged into three zones. These chondrocytes mediate linear bone growth through synchronized proliferation, differentiation, and production of matrix. LÄS MER
4. Influences of in vivo and in vitro loading on the proteoglycan synthesis of articular cartilage chondrocytes
Sammanfattning : In this study, the biosynthesis of proteoglycans (PGs) was examined in articular cartilage of canine hip joint after long-distance running experiment and in bovine chondrocyte cultures during in vitro loading with hydrostatic pressure. In addition, new assays were developed for more sensitive quantitation of glycosaminoglycans (GAGs) and PGs. LÄS MER
5. Inflammatory response in equine joints : studies on proinflammatory cytokines in diseased joints and chondrocyte cultures
Sammanfattning : Proinflammatory cytokines mediate inflammatory responses as well as regulate tissue metabolism. Thus, they may provide a link between inflammation and other pathologic findings seen in equine joint disease. LÄS MER
