1. Disease-causing Keratin Mutations and Cytoskeletal Dysfunction in Human Skin : In vitro Models and new Pharmacologic Strategies for Treating Epidermolytic Genodermatoses
Sammanfattning : Epidermolysis bullosa simplex (EBS) and epidermolytic ichthyosis (EI) are rare skin fragility diseases characterized by intra-epidermal blistering due to autosomal dominant-negative mutations in basal (KRT5 or KRT14) and suprabasal (KRT1 or KRT10) keratin genes, respectively. Despite vast knowledge in the disease pathogenesis, the pathomechanisms are not fully understood, and no effective remedies exist. LÄS MER
2. On keratin mutations in epidermolytic hyperkeratosis and the regulation of keratin expression by retinoids
Sammanfattning : Epidermolytic hyperkeratosis is a rare inherited disease of the skin caused by a dominant-negative mutation in keratin 1 (K1) or 10 (K10). Keratins are the major structural protein in epidermis and mutations causes instability of intermediate filament and keratinocyte fragility. LÄS MER
3. Bioprocessing of Recalcitrant Substrates for Biogas Production
Sammanfattning : The application of anaerobic digestion (AD) as a sustainable waste management technology is growing worldwide, due to high energy prices as well as increasingly strict environmental regulations. The growth of the AD industry necessitates exploring new substrates for their utilisation in AD processes. LÄS MER
4. Construction, expression and evaluation of anti-keratin 8 single-chain antibody fragments
Sammanfattning : Antibodies are glycoproteins specifically binding to a variety of antigens and today extensively used in prevention, diagnosis and treatment of diseases. Carcinomas originate from epithelial tissues and are the most common forms of human malignancies. In the necrotic areas of carcinomas significant amounts of keratins (K) are found extracellularly. 5. Foreign molecules in biomembranes : Molecular effects on intact stratum corneum and model lipid systems
Sammanfattning : This thesis mainly explores how the molecular mobility of lipid and protein components in the outermost layer of the skin, the stratum corneum (SC), varies with different conditions and upon adding foreign compounds. These studies require methods that makes it possible to detect and characterize the minor amount of fluid components in the highly ordered solid SC sample with resolved molecular information. LÄS MER
