  1. 1. High Content Analysis of Proteins and Protein Interactions by Proximity Ligation

    Författare :Karl-Johan Leuchowius; Ola Söderberg; Ulf Landegren; Paul Goldsmith; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; in situ; proximity ligation; flow cytometry; high content screening; rolling circle amplification; in situ PLA; single-cell; single-molecule; protein interactions; drug screening; post-translational modifications; Molecular medicine; Molekylär medicin; Molekylär medicin; Molecular Medicine;

    Sammanfattning : Fundamental to all biological processes is the interplay between biomolecules such as proteins and nucleic acids. Studies of interactions should therefore be more informative than mere detection of expressed proteins. Preferably, such studies should be performed in material that is as biologically and clinically relevant as possible, i.e. LÄS MER

  2. 2. Visualization of Protein Activity Status in situ Using Proximity Ligation Assays

    Sammanfattning : In 2001 the human proteome organization (HUPO) was created with the ambition to identify and characterize all proteins encoded in the human genome according to several criteria; their expression levels in different tissues and under different conditions; the sub-cellular localization; post-translational modifications; interactions, and if possible also the relationship between their structure and function.When the knowledge of different proteins and their potential interactions increases, so does the need for methods able to unravel the nature of molecular processes in cells and organized tissues, and ultimately for clinical use in samples obtained from patients. 3. 3. Characterizing phenotypes of Mycobacterium tuberculosis and exploring anti-mycobacterial compounds through high content screening

    Författare :Sadaf Kalsum; Maria Lerm; Thomas Lundbäck; Annika Jenmalm; Lena Serrander; Priscille Bordin; Linköpings universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Sammanfattning : Tuberculosis (TB), an airborne disease and one of the top 10 causes of death globally, is caused by Mycobacterium tuberculosis (Mtb). Current standard therapy for TB treatment includes multiple drugs for a period of at least 6 months. LÄS MER

    Sammanfattning : This thesis presents automatic image and data analysis methods to facilitate and improve microscopy-based research and diagnosis. New technologies and computational tools are necessary for handling the ever-growing amounts of data produced in life science. LÄS MER

  5. 5. Towards High-Throughput Phenotypic and Systemic Profiling of in vitro Growing Cell Populations using Label-Free Microscopy and Spectroscopy : Applications in Cancer Pharmacology

    Författare :Obaid Aftab; Mats Gustafsson; Neil Carragher; Uppsala universitet; []
    Nyckelord :label free vesicle detector; high-throughput; phase contrast microscopy; Library of Pharmacologically Active Compounds; High Content Screening; fluorometric microculture cytotoxicity assay; nuclear magnetic resonance; mass spectrometry;

    Sammanfattning : Modern techniques like automated microscopy and spectroscopy now make it possible to study quantitatively, across multiple phenotypic and molecular parameters, how cell populations are affected by different treatments and/or environmental disturbances. As the technology development at the instrument level often is ahead of the data analytical tools and the scientific questions, there is a large and growing need for computational algorithms enabling desired data analysis.
