    Sammanfattning : The environmental pollutant 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induces thymus atrophy and immunosuppression in all animal species examined and these effects constitute a potential risk to humans. TCDD can via binding to the intracellular aryl hydrocarbon receptor (AhR), modulate transcription of a number of genes and the aim of this thesis has been to identify such genes, which may explain the mechanisms behind TCDD-mediated immunotoxicity.     Sammanfattning : Polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs), two groups of struc-turally related chlorinated aromatic hydrocarbons, generally referred to as “dioxins” are of great concern due to their extreme toxicity and presence in all compartments of the environment. Since they occur at very low levels, their analysis is complex and challenging, and there is a need for cost-efficient, reliable and rapid analytical alternatives to the expensive methods in-volving use of gas chromatography high-resolution mass spectrometry (GC-HRMS).     Sammanfattning : In mammals the basic helix-loop-helix/Per-Arnt-Sim (bHLH/PAS) family of transcription factors includes the dioxin receptor which is a ligand-dependent transcription factor which mediates biological effects of environmental pollutants such as 2,3,7,8-tetrachlorodibenzo-rho-dioxin (dioxin). In the absence of ligand, the dioxin receptor is associated with the molecular chaperone Hsp90 and two Hsp90-interacting proteins, the co-chaperone p23 and an immunophilin-like protein XAP2. LÄS MER

    Sammanfattning : The intracellular dioxin receptor is a bHLH/PAS protein which mediates signal transduction by dioxin and upon heterodimerisation with the bHLH/PAS partner factor Arnt, functions as a ligand-activated transcription factor by binding to specific xenobiotic response elements in promoters of target genes. Although the physiological function of the dioxin receptor and the nature of a possible physiological ligand are presently not known, targeted disruption of the mouse dioxin receptor gene has indicated that the receptor may play an important role in hepatic development.
