    Sammanfattning : Covalent bonds between lignin and carbohydrates, forming a matrix referred to as lignin carbohydrate complexes (LCC), remain one of the most controversial topics in wood chemistry. A key issue is whether they are formed during chemical and mechanical pretreatments of the compact wood structure or actually present in wood prior to isolation. LÄS MER

  2. 2. Design and Synthesis of Serine and Aspartic Protease Inhibitors

    Författare :Fredrik Wångsell; Ingemar Kvarnström; Olle Karlsson; Linköpings universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; organic chemistry; organic synthesis; metathesis; HCV; NS3; protease; proline isosteres; inhibitor; aspartic protease inhibitors; hydroxyethylene; Organic synthesis; Organisk syntes;

    Sammanfattning : This thesis describes the design and synthesis of compounds that areintended to inhibit serine and aspartic proteases. The first part of the text deals with preparation of inhibitors of the hepatitis C virus (HCV) NS3 serine protease. Hepatitis C is predominantly a chronic disease that afflicts about 170 million people worldwide. LÄS MER

  3. 3. Structure-Based Design and Synthesis of Protease Inhibitors Using Cycloalkenes as Proline Bioisosteres and Combinatorial Syntheses of a Targeted Library

    Författare :Fredrik Thorstensson; Ingemar Kvarnström; Charles Hedgecock; Linköpings universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; organic chemistry; organic synthesis; combinatorial synthesis; metathesis; olefin metathesis; thrombin; HCV; NS3; protease; proline isosteres; inhibitor; Organic synthesis; Organisk syntes;

    Sammanfattning : Structure-based drug design and combinatorial chemistry play important roles in the search for new drugs, and both these elements of medicinal chemistry were included in the present studies. This thesis outlines the synthesis of protease inhibitors against thrombin and the HCV NS3 protease, as well as the synthesis of a combinatorial library using solid phase chemistry. LÄS MER

  4. 4. Conformations of Flexible Oligosaccharides : Molecular Simulations and NMR spectroscopy

    Författare :Robert Pendrill; Göran Widmalm; Jens Duus; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Carbohydrates; Oligosaccharide; Conformation; NMR spectroscopy; MD simulations; organisk kemi; Organic Chemistry;

    Sammanfattning : The conformational preferences of several oligosaccharides are investigated herein using a combination of NMR spectroscopy and molecular dynamics (MD) simulations, focusing on the torsion angles associated with the glycosidic linkages.Strategies for obtaining usable J-HMBC spectra for carbons with an adjacent 13C label are described. 5. 5. Conformational studies of carbohydrates: MD simulation and NMR spectroscopy

    Författare :Robert Pendrill; Lena Mäler; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : This thesis describes various aspects of conformational studies of carbohydrates, from development of the methods by which experimental parameters are gathered to the application of NMR spectroscopy and MD simulation for the analysis of a disaccharide. Paper I describes the use of site-specific 13C labeling as a tool to resolve spectral overlap of 1H frequencies in a trisaccharide, allowing the measurement of important crossrelaxation rates and long-range couplings which were previously obscured.
