11. Blood-Brain Barrier Transport : Investigation of Active Efflux using Positron Emission Tomography and Modelling Studies
Sammanfattning : This thesis examines the transport of exogenous molecules across the blood-brain barrier (BBB), focusing on active efflux, using positron emission tomography (PET), computer simulation and modelling. P-glycoprotein (P-gp) inhibition was studied using [11C]verapamil and [11C]hydroxyurea was investigated as a new marker for active efflux transport. LÄS MER
12. Pharmacology and resistance mechanisms of nucleoside analogues and topoisomerase II interactive agents : studies on human leukemia cells with a focus on cross-resistance
Sammanfattning : The purpose of this thesis was to elucidate mechanisms of action and resistance of clinically relevant nucleoside analogues and topoisomerase interactive agents in human leukemia cell lines and leukernia cells isolated from peripheral blood of leukemia patients. Interactions and cross-resistance patterns of these different cytotoxic drug families were also studied since these drugs are usually administrated in combination in the clinic. LÄS MER
13. Pericyte Influence on Drug Delivery Across the Blood-Brain Barrier : Implications for Therapy of Neurodegenerative Diseases
Sammanfattning : The blood-brain barrier (BBB) represents a complex interface between the brain parenchyma and systemic blood circulation, strictly controlling exchange of substances between the two sites. Pericytes are mural cells located on the abluminal membrane of the brain endothelium, involved in BBB formation and maintenance. LÄS MER
14. In Vivo Active Drug Uptake and Efflux at the Blood-Brain Barrier : With Focus on Drug Transport Interactions
Sammanfattning : The blood-brain barrier (BBB) controls the movement of substances into and out of the brain. The tight junctions between endothelial cells and energy dependent transporters in the BBB influence rate and extent of drug distribution to the brain. LÄS MER
15. Pharmacological properties of radiotracers that measure p-glycoprotein function and density
Sammanfattning : Energy-dependent transporters of the ATP-binding cassette (ABC) family regulate the movement of molecules across cellular membranes. Several of these transporters are expressed in the endothelial cells of brain microvessels (blood-brain barrier) to protect brain tissue from exposure to toxins in the blood. LÄS MER
