1. Structure and function of the cytoskeleton in cardiac and skeletal muscle
Sammanfattning : We have examined the functional and structural roles of the cytoskeletal protein desmin in cardiac and skeletal muscles using a genetically modified mouse (Des-/-) with the desmin gene ablated. Desmin forms filaments at the Z-disks in the striated muscle sarcomere, have connections to the sarcolemma and most likely align sarcomeres and whole cells. LÄS MER
2. Regulatory Functions of the Juxtaglomerular Apparatus
Sammanfattning : The tubuloglomerular feedback mechanism is an important regulator in the juxtaglomerular apparatus and it detects flow dependent alterations in luminal NaCl concentration ([NaCl]) at the macula densa (MD) cell site via a Na+-K+-2Cl cotransporter. Signals are sent by the MD to adjust the afferent arteriole tone and altering release of renin. LÄS MER
3. Genetic studies on cardiovascular disease - identification of novel drug targets
Sammanfattning : The aim of this thesis was to identify novel potential drug targets of cardiovascular disease with a focus on G-protein coupled receptors. We identified a common genetic variant of the ATP receptor P2Y11, where an alanine is substituted with a threonine at position 87 of the P2Y11 receptor that increases the risk of developing acute myocardial infarction. 4. Modulation of Receptor Signaling and Functional Selectivity in Neutrophils
Sammanfattning : Neutrophils are important effector cells of the innate immune system and in the regulation of inflammation. Many of their functions, such as chemotactic migration, secretion of granule constituents and activation of the oxygen radical-producing NADPH-oxidase, are regulated by cell surface receptors. 5. Adenosine A2A and ATP receptors in PC12 cells
Sammanfattning : PC12 cells, originally isolated from a rat pheochromocytoma, are widely used to study receptor pharmacology and nerve cell differentiation. They are the neoplastic counterpart of adrenal chromaffin cells, but upon treatment with NGF they acquire a neuronal phenotype. LÄS MER
