    Sammanfattning : The aim of this thesis was to identify novel potential drug targets of cardiovascular disease with a focus on G-protein coupled receptors. We identified a common genetic variant of the ATP receptor P2Y11, where an alanine is substituted with a threonine at position 87 of the P2Y11 receptor that increases the risk of developing acute myocardial infarction.     Sammanfattning : The aims of this dissertation were to investigate the effects of extracellular purines on insulin secretion, and apoptosis in mouse pancreatic islets and β-cells; to examine if high glucose and free fatty acids induces β-cell apoptosis via autocrine effects of ADP acting on the P2Y13 receptor; and to investigate the modulation of extracellular purines in vascular smooth muscle cells (VSMCs) through ectonucleotidase or ATP release under the effect of high glucose. The expression of the ADP receptors P2Y1 and P2Y13 were shown in both mouse pancreatic islets and isolated β-cells using real-time PCR quantification. LÄS MER

    Sammanfattning : The tubuloglomerular feedback mechanism is an important regulator in the juxtaglomerular apparatus and it detects flow dependent alterations in luminal NaCl concentration ([NaCl]) at the macula densa (MD) cell site via a Na+-K+-2Cl cotransporter. Signals are sent by the MD to adjust the afferent arteriole tone and altering release of renin. LÄS MER
