  1. 6. Blood-Brain Barrier Transport : Investigation of Active Efflux using Positron Emission Tomography and Modelling Studies

    Författare :Stina Syvänen; Margareta Hammarlund-Udenaes; Mats Bergström; Gunnar Blomquist; Jashvant Unadkat; Uppsala universitet; []
    Nyckelord :Pharmaceutical biosciences; pharmacokinetics; P-glycoprotein; blood-brain barrier; modelling; PET; active efflux; species differences; [11C]verapamil; drug development; Farmaceutisk biovetenskap;

    Sammanfattning : This thesis examines the transport of exogenous molecules across the blood-brain barrier (BBB), focusing on active efflux, using positron emission tomography (PET), computer simulation and modelling. P-glycoprotein (P-gp) inhibition was studied using [11C]verapamil and [11C]hydroxyurea was investigated as a new marker for active efflux transport. LÄS MER

  2. 7. Neural Control of Movement : Motor Neuron Subtypes, Proprioception and Recurrent Inhibition

    Författare :Anders Enjin; Klas Kullander; Samuel L. Pfaff; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; motor neuron; proprioception; recurrent inhibition; molecular marker; Ia afferent; development; transgenic mice; Renshaw cell; Neurobiology; Neurobiologi; Neuroscience; Neurovetenskap;

    Sammanfattning : Movement is central for life, and all animals depend on accurate regulation of movement for purposeful behavior. There is great diversity of movements, ranging between simple and vital breathing movements to minute and subtle movements of the face used to communicate emotions. LÄS MER

  3. 8. ATP-Binding Cassette Efflux Transporters and Passive Membrane Permeability in Drug Absorption and Disposition

    Författare :Pär Matsson; Per Artursson; Ulf Norinder; Peter Swaan; Uppsala universitet; []
    Nyckelord :Pharmaceutics; ATP-binding cassette; ABC transporter; P-gp; P-glycoprotein; ABCB1; BCRP; Breast cancer resistance protein; ABCG2; MRP2; Multidrug-resistance associated protein 2; ABCC2; Membrane permeability; Drug transport; Active transport; Passive diffusion; Multivariate data analysis; PLS; OPLS; QSAR; Galenisk farmaci;

    Sammanfattning : Transport into and across the cells of the human body is a prerequisite for the pharmacological action of drugs. Passive membrane permeability and active transport mechanisms are major determinants of the intestinal absorption of drugs, as well as of the distribution to target tissues and the subsequent metabolism and excretion from the body. Sammanfattning :    With the advancement and accessibility of local probing techniques that can operate at the submicron scale it has become possible to analyse the local corrosion properties of industrially important metallic materials and relate these properties to microstructure characteristics of the same materials. In this doctoral study the focus has been on copper-zinc samples, both as industrial brass alloys and as micro-patterned copper-zinc samples. LÄS MER

  5. 10. In vitro and in silico Predictions of Hepatic Transporter-Mediated Drug Clearance and Drug-Drug Interactions in vivo

    Författare :Anna Vildhede; Per Artursson; Maria Karlgren; Maria Kjellsson; Ulf Norinder; Kim Brouwer; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; OATP1B1; OATP1B3; OATP2B1; NTCP; drug transporters; human hepatocytes; atorvastatin; pitavastatin; proteomics; sandwich-cultured human hepatocytes; SCHH; mechanistic modeling; in vitro-in vivo extrapolation; transport inhibition; hepatic uptake; hepatocyte isolation; transporter contribution; Biopharmaceutics; Biofarmaci;

    Sammanfattning : The liver is the major detoxifying organ, clearing the blood from drugs and other xenobiotics. The extent of hepatic clearance (CL) determines drug exposure and hence, the efficacy and toxicity associated with exposure. Drug-drug interactions (DDIs) that alter the hepatic CL may cause more or less severe outcomes, such as adverse drug reactions. LÄS MER
