1. Characterization of two novel fatty acid dioxygenases : Linoleate diol synthase and manganese lipoxygenase
Sammanfattning : Two classes of fatty acid dioxygenases, PGH synthases (hemeproteins) and lipoxygenases (non-heme iron proteins), are of eminent pharmacological importance. This thesis describes a PGH synthase-related dioxygenase designated linoleate diol synthase (LDS) and the first native occurring manganese-containing lipoxygenase. LÄS MER
2. Asymmetric transformation of ß- and γ-functionalized alcohols : Study of combined ruthenium-catalyzed racemization and enzymatic resolution
Sammanfattning : The major part of this thesis describes the asymmetric synthesis of β- and γ-amino alcohols through the combination of ruthenium catalyzed racemization and enzymatic kinetic resolution. The dynamic kinetic resolution, DKR, protocol for chlorohydrins was improved by employing Bäckvall’s catalyst, which is a base activated racemization catalyst, in combination with Burkholderia cepacia lipase. 3. Outer Coordination Spheres Engineering of Ru-based Molecular Water Oxidation Catalysts
Sammanfattning : The outer coordination sphere plays a vital role in metalloenzyme catalysis, while this principle is relatively less considered in the design of man-made molecular catalysts. This thesis investigates the role of outer coordination spheres in water oxidation by a series of ruthenium-based complexes. LÄS MER
4. Evolution and structure of leukotriene A4 hydrolase
Sammanfattning : Leukotriene (LT) B4 is a potent leukocyte chemotactic and aggregating agent involved in a variety of inflammatory and allergic disorders. The enzyme leukotriene A4 hydrolase catalyzes the hydrolysis of LTA4 into this proinflammatory mediator, leukotriene B4. LÄS MER
5. Push the limitations of crystal structure determination by 3D electron diffraction : From inorganic porous materials to biomolecules
Sammanfattning : Structure elucidation is fundamental to understanding the chemical and physical properties of a material. Three-dimensional electron diffraction (3D ED) has shown great power for structure determination of nanometer- or submicrometer-sized crystals that are either too small or too complex for X-ray diffraction.
