1. Studies on hereditary C2 deficiency: Frequent occurrence of severe infections, atherosclerosis and rheumatological manifestations
Sammanfattning : The complement system is a part of the innate immunity and is essential in the defence against microorganisms. Hereditary C2 deficiency (C2D) is one of the most common complement deficiency states with an estimated prevalence of 1:20,000 in persons of Western descent. LÄS MER
2. Molecular dissection of Bruton’s tyrosine kinase signaling in hematopoietic cells using RNAi
Sammanfattning : RNA mediated gene silencing, RNA interference (RNAi), has recently been identified in mammalians. This thesis describes the utilization and development of RNAi based tools for signal transduction research. The effects of Bruton s tyrosine kinase (Btk) down-regulation with RNAi have been studied with gene expression profiling in hematopoietic cells. LÄS MER
3. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development
Sammanfattning : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 4. Multidisciplinary analysis of HIV-1 elite controllers
Sammanfattning : Chronic HIV-1 infection is characterized by progressive depletion of CD4+ T lymphocytes, persistent immune activation and ongoing viral replication, leading to a profound immunodeficiency state if left untreated with antiretroviral therapy. However, a small percentage of infected individuals are able to maintain durable control of HIV replication and stable CD4 counts, in the absence of antiretroviral treatment (ART). LÄS MER
5. Innate immunity in the immunopathogenesis of chronic viral infection
Sammanfattning : Natural killer (NK) cells have a key role in control and clearance of viral infections. To carry out this function NK cells are capable of recognising infected cells and responding with induction of apoptosis in these cells, and production of pro-inflammatory cytokines. LÄS MER
