1. Magnetic relaxation studies of self-associating and membrane proteins
Sammanfattning : Magnetic relaxation dispersion measurements have been performed on aqueous protein solutions to study the dynamics of waters in the proton transport channel of bacteriorhodopsin and the self-association of the proteins BPTI, lysozyme and beta-lactoglobulin. The measurements have focused on the bulk relaxation rates, R1 and R2, of three water nuclei 1H, 2H and 17O that are coupled to the protein environments by exchange and therefore report on properties such as the protein tumbling and dynamics of internal waters. 2. Structure, dynamics and interactions of biomolecules : Investigations by NMR spectroscopy and computational methods
Sammanfattning : In this thesis, the structure, dynamics and interactions of proteins and carbohydrates are investigated using mainly NMR spectroscopy and computer simulations. Oligosaccharides representing a Salmonella O-antigen have been synthesized and their dynamic behavior and interaction with the bacteriophage P22 tail-spike protein have been studied by NMR spectroscopy, MD and docking simulations. 3. Phage–derived Endolysins as Potential Antibacterials : A Study of Peptidoglycan Hydrolase and Mycolylarabinogalactan Esterase Enzymes
Sammanfattning : Bacteriophages, or phages, are viruses that infect bacteria, at the end of their life cycle produce a set of enzymes called endolysins to lyse host cells from within, facilitating the release of the viral progeny. Due to their lytic activity, endolysins have gained great interest as potential antibacterials targeting both Gram–positive and –negative bacteria, especially in the actual context of increasing rates of antibiotics resistance.
