  1. 1. Improvements of the Bromodeoxyuridine-DNA Flow Cytometry Method for the Study of Cell Proliferation

    Författare :Maria C Johansson; Bröstcancer-genetik; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; head and neck cancer; [3H]thymidine; energy transfer; cell cycle; xenografted tumours; cultured cells; potential doubling time; labelling index; DNA synthesis time; cell proliferation; growth kinetics; Bromodeoxyuridine; DNA flow cytometry; cancer; onkologi; Cytologi; cancerology; oncology; Cytology;

    Sammanfattning : Potential doubling time (Tpot), DNA synthesis time (TS), and labelling index (LI) are fundamental growth kinetics parameters in clinical and experimental cancer research, which may be of further practical importance regarding prognosis and treatment prediction of cancer. They can be measured by bromodeoxyuridine(BrdUrd)/flow cytometry (FCM) methods, where BrdUrd, an analogue of thymidine, is incorporated into DNA and quantified simultaneously with the DNA content. 2. 2. Neurorestorative strategies involving neurogenesis, neuronal precursors and stem cells in animal models of Parkinson's disease

    Författare :Ming Zhao; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Parkinson s disease; substantia nigra pars compacta; adult neurogenesis; stereology; apoptosis; dopamine; tyrosine hydroxylase; nestin; 5-bromodeoxyuridine; [3H]-thymidine; MPTP; 6-OHDA; platelet-derived growth factor-BB;

    Sammanfattning : The general aims of the thesis is to provide evidence for neurogenesis in the adult substantia nigra pars compacta (SNpc), where the dopamine-producing neurons lost in Parkinson's disease (PD) reside; to optimize methods used for detecting newborn nerve cells in adult brain regions with a low rate of neurogenesis; to explore the mechanism of nerve cell death in animal models of parkinsonism and early degenerative and restorative changes in nigral cell populations; and to develop a therapeutic approach that could translate into a future restorative disease-modifying strategy in PD. 3. 3. HIV-1 reverse transcriptase activity assays based on monoclonal antibodies to native enzyme : Applications on studies of antiviral substances and immunological variation of reverse transcriptase

    Författare :Ann-Sofie Rytting; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Genetics; HIV-1; reverse transcriptase; monoclonal antibody; activity assay; subtype; RT inhibitor; Genetik; Clinical genetics; Klinisk genetik; Medical Genetics; medicinsk genetik;

    Sammanfattning : Specific detection of HIV-1 reverse transcriptase (RT) activity in crude materials was enhanced using a capture assay for immunopurification and subsequent activity assay. The purification step was achieved by anti-HIV-1 RT monoclonal antibodies (Mabs) immobilized onto a solid carrier. LÄS MER

  4. 4. Exploring the metagenome of the Baltic Sea sediment

    Författare :Fredrik Hårdeman; Sara Sjöling; Gunnar Sandström; Kornelia Smalla; Södertörns högskola; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Maringeologi; Bakterier;

    Sammanfattning : Environmental microorganisms are fundamental to ecosystem function, acting as drivers in processes such as primary production, organic matter remineralisation, pollution remediation and global biogeochemical cycling. However, the study of the bacterial communities requires the application of advanced culture-independent methods considering that only a small fraction of the community is otherwise accessed. 5. 5. Neurogenesis and gliogenesis after focal brain ischemia. Modulation by enriched environment and exercise

    Författare :Mila Komitova; Göteborgs universitet; []
    Nyckelord :astrocyte; hippocampus; neural progenitor cells; neural stem cells; neurogenesis; neurotrophins; rat; regeneration; rehabilitation; stroke;

    Sammanfattning : New neurons are formed from dividing neural stem cells in restricted areas in the adult mammalian brain: the dentate gyrus of the hippocampal formation (DG) and the forebrain subventricular zone (SVZ). New glia also form constitutively and seem to be important for neuronal function. LÄS MER
