  1. 1. Enantioselective drug metabolism catalysed by the polymorphic CYP2D6 and CYP2C19 : a methodological investigation focused on mianserin, mephenytoin and omeprazole

    Författare :Gunnel Tybring; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Drug metabolism; CYP2D6; CYP2CI9; mianserin; mephenytoin; omeprazole; enantiomers; chiral analysis;

    Sammanfattning : The aim of this thesis was to develop enantioselective analytical methods and apply them to studies of oxidative metabolism of chiral drugs catalysed by the polymorphic cytochrome P450 CYP2D6 and CYP2C 19 enzymes. The work has been focused on mianserin, mephenytoin and omeprazole. LÄS MER

  2. 2. CYP2C19 catalyzed drug metabolism in different populations

    Författare :Karin Herrlin; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :CYP2C19; drug metabolism; cytochrome P450; mephenytoin; omeprazole; proguanil chloroguanide ; citalopram; debrisoquine; CYP2D6; gene duplication; pharmacogenetics;

    Sammanfattning : Metabolism catalyzed by the cytochrome P450 enzyme CYP2C19 varies between individuals, who can be either efficient (EM) or poor metabolizers (PM). In this thesis the metabolism of four different CYP2C19 substrates (citalopram, mephenytoin, omeprazole, and proguanil) was investigated in vivo in Africans (Ethiopians an Tanzanians) an in Swedish is Caucasians. 3. 3. Pharmacokinetic drug-drug interactions in the management of malaria, HIV and tuberculosis

    Författare :Doaa Elsherbiny; Ulrika Simonsson; Mats Karlsson; Lars Ståhle; Uppsala universitet; []
    Nyckelord :Pharmacokinetics Pharmacotherapy; Pharmacokinetics; Drug-drug interactions; Cytochrome P-450; Artemisinin antimalarials; Nevirapine; Lopinavir; Rifampicin; NONMEM; Farmakokinetik Farmakoterapi;

    Sammanfattning : Malaria, Human Immunodeficiency Virus (HIV) and tuberculosis (TB) are global health problems having their worst situation in sub-Saharan Africa. Consequently, concomitant use of antimalarial, antiretroviral and antitubercular drugs may be needed, resulting in a potential risk of drug-drug interactions. LÄS MER

  4. 4. CYP2C19 polymorphisms in drug metabolism and response

    Författare :Ming Chang; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Individuals vary widely in their response to drug treatment. After receiving doses of a drug that are recommended based on a population average, some patients could have an insufficient response, whereas others may experience adverse effects. LÄS MER

  5. 5. Genetic polymorphism of xenobiotic metabolising enzymes : implications for interindividual and interethnic differences in drug metabolism and cancer susceptibility

    Författare :Irene Persson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Lung cancer; drug metabolism; genetic polymorphism; cytochrome P450; CYPIAI; CYP2EI; CYP2D6; interethnic differences.;

    Sammanfattning : Genetic factors are of importance for interindividual and interethnic differences in drug response and cancer susceptibility. In the present investigation the role of genetic polymorphism in the cytochromes P450 CYPIAI, CYP2C19, CYP2D6, CYP2EI as well as in glutathione S-transferase Ml (GSTMI) and microsomal epoxide hydrolase (mEH) for differences in drug response and cancer susceptibility has been investigated in various ethnic populations.
