1. Automating STED microscopy for functional and structural live-cell imaging
Sammanfattning : Optical microscopy imaging methods are today invaluable tools for studies in life sciences as they allow visualization of biological systems, tissues, cells, and sub-cellular compartments from millimetres down to nanometres. The invention and development of nanoscopy in the past 20 years has pushed fluorescence microscopy down to the nanoscale, reaching beyond the natural diffraction limit of light that does not allow focusing of visible light below sizes of around 200 nm, and into the realm of what was previously only thought possible with electron microscopy. 2. Development of quantitative optical microscopy methods for single cell analysis
Sammanfattning : Optical microscopy has brought powerful methods to biological research, in particular the possibility to assay dynamic molecular processes within living individual cells. This thesis deals with various aspects of fluorescence and Raman microscopy, such as the possibility of avoiding cellular damage or stress caused by illumination, how to manipulate single cells for spectroscopic analysis and how to quantify images automatically. 3. Mechanisms regulating Natural Killer Cell Cytotoxicity
Sammanfattning : Over the last 50 years, cancer survival rates have steadily improved thanks to earlier detection and novel treatment regimens. For example, drugs that act on the immune system, so-called immunotherapy, have drastically increased the prospect of survival for patients suffering from some of the most aggressive cancer types including advanced malignant melanoma. 4. Detection and quantification of lipid-mediated siRNA delivery to the cytosol
Sammanfattning : Cancer is one of the leading causes of death globally. Novel therapeutics is urgently needed to advance treatment, especially for generalized disease where current treatment options have a poor prognosis. In particular, targeted therapy that can address genetic changes would be of great value. LÄS MER
5. Advancing DNA-based proximity methods
Sammanfattning : Cellular functions are governed by intricate chains of interactions between proteins. In order to properly understand cellular biology one must look into, not only protein function, but also its interacting network. LÄS MER
