1. Chromatin, histones, and epigenetic tags
Sammanfattning : The fundamental building blocks of chromatin are the nucleosomes. Each such unit is composed of about 200 bp of DNA, the well-conserved core histones (H2A, H2B, H3 and H4) and a linker histone (H1). LÄS MER
2. Antigen interaction with B cells in two proliferative disorders : CLL and MGUS
Sammanfattning : The aim of the work presented in this thesis was to elucidate B cell interaction with antigen in the two B cell proliferative disorders chronic lymphocytic leukemia (CLL) and monoclonal gammopathy of undetermined significance (MGUS). In the first part we investigated the antigen specificity of CLL cells and characterized Epstein-Barr virus (EBV)-transformed CLL cell lines with regard to phenotype and genotype. LÄS MER
3. Restricted antigen recognition in B cell chronic lymphocytic leukemia
Sammanfattning : Chronic lymphocytic leukemia (CLL) cells are considered to be derived from antigen-exposed B cells. To further explore the antigen-driven selection behind the leukemogenesis of CLL, we performed immunoglobulin (Ig) specificity screening of 7 CLL cell lines and 23 primary CLL clones from patient peripheral blood. 4. Genome-wide epigenetic profiling of B cell leukemia and lymphoma
Sammanfattning : Epigenetic modifications, at the level of DNA methylation and post-translational modifications of histone tails cooperatively function in the organization of the genome, and thereby establish the gene expression profiles, phenotypes, and cellular fates. In this work, we investigated the aberrant epigenome in chronic lymphocytic leukemia (CLL) which is one of the most frequent lymphoid malignancies in the west including the Nordic countries. LÄS MER
5. Soluble intercellular and vascular cell adhesion molecules-1 in lymphoid neoplasms : A clinical and prognostic study of Hodgkin's disease, non-Hodgkin's lymphomas and chronic B-lymphocytic leukaemia
Sammanfattning : The interaction of soluble adhesion molecules with adhesion ligands expressed by immunological cells interfere with the cell-cell adhesion essential for the signal transductions that initiate immunological responses. Preliminary data suggest that soluble adhesion molecules also promote angiogenesis. LÄS MER
