1. Thrombin generation in different cohorts : Evaluation of the haemostatic potential
Sammanfattning : The aim of this thesis is to evaluate thrombin generation in patients with thrombophilia (Paper I), in patients with venous thromboembolism (Paper II), in healthy women during the menstrual cycle (Paper III), in patients with liver disease (Paper IV) and in patients with mild deficiency of factor VII (Paper V).For this purpose, thrombin generation was measured in platelet poor plasma by the calibrated automated thrombogram (CAT®) assay. 2. Exploring Anti-FVIII Antibodies in Haemophilia A - Role in In Vitro Haemostasis and Clinical Disease
Sammanfattning : Haemophilia A (HA) is caused by defective synthesis of coagulation factor VIII(FVIII), which has serious effects on haemostasis; joints being the most common site of bleeding. The development of FVIII replacements has improved the situation for patients with haemophilia such that chronic arthropathy can be prevented, and life expectancy and the quality of life have increased. 3. Thrombin activatable fibrinolysis inhibitor (TAFI) in different hemorrhagic and thrombotic conditions
Sammanfattning : Until recently the coagulation and fibrinolytic systems were usually considered as separate entities. Accounts of a fibrinolysis inhibitor that is generated by thrombin shed more light on this field and changed the concept by giving thrombin, the pivotal enzyme in hemostasis, another important role in the down-regulation of fibrinolysis. 4. Novel chemical and enzymatic routes to the generation of heparin- related polysaccarides
Sammanfattning : Heparin and heparan sulphate (HS) are structurally related polysaccharides influencing various biological processes. The biological functions of heparin/HS largely depend on interactions between the negatively charged polysaccharide chains and a variety of proteins. LÄS MER
5. Determination of the overall haemostasis potential and fibrin gel permeability : method development and application in research and in clinical materials
Sammanfattning : We have previously developed a laboratory method that may screen the Overall Haemostasis Potential (OHP) in plasma based on spectophotometric measurement of an area under the fibrin aggregation curve in citrated plasma samples to which tiny amounts of exogenous thrombin and tissue-plasminogen activator (t-PA) are added. When the fibrin aggregation curve is created, the fibrinogen originally present in plasma is gradually converted to fibrin by the generated thrombin. LÄS MER
