  1. 1. Strain- and bioprocess-design strategies to increase production of (R)-3-hydroxybutyrate by Escherichia coli

    Författare :Mónica Guevara-Martínez; Gen Larsson; Antonius J. A. van Maris; Pau Ferrer; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; E. coli; R -3-hydroxybutyrate; metabolic engineering; bioprocess design; NADPH; acetic acid; thioesterase; BAC; Bioteknologi; Biotechnology;

    Sammanfattning : Microbial bio-based processes have emerged as an alternative to replace fossil-based processes for the production of fuels and chemicals. (R)-3-hydroxybutyrate (3HB) is a medium-value chemical that has gained special attention as a precursor of antibiotics and vitamins, as a monomer for the synthesis of tailor-made polyesters and as a nutritional source for eukaryotic cells. 2. 2. Molecular cloning and characterization of the murine acyl-CoA thioesterase CTE-I

    Författare :Per J G Lindquist; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Acyl-CoA thioesterase; acyl-CoA; peroxisome proliferator-activated receptor alpha; fatty acid metabolism;

    Sammanfattning : Mouse cytosolic type I acyl-CoA thioesterase (CTE-I) is a member of the type I family of acylCoA thioesterases. These enzymes hydrolyze acyl-CoAs to non-esterified fatty acids (NEFAs) and Coenzyme A (CoA-SH) and are strongly induced in rodent liver by a diverse group of compounds called peroxisome proliferators. 3. 3. The type-I acyl-CoA thioesterase/acyltransferase gene family : linking structure to function

    Författare :James O'Byrne; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Type-I acyl-CoA thioesterase acyltransferase gene family; structure function relationship; nueleophilic elbow; esterase transferase activity;

    Sammanfattning : Acyl-CoA thioesterases hydrolyse acyl-CoAs to non-esterified fatty acids and coenzyme A (CoASH), thereby regulating intracellular levels of these compounds. We have recently identified and initiated characterisation of a novel, unique gene family of acyl-CoA thioesterases/acyltransferases, called the Type-I acyl-CoA thioesterase/acyltransferase gene family. 4. 4. Characterization of peroxisome proliferator-activated receptor alpha (PPARa) regulated acyl-CoA thioesterases involved in lipid metabolism

    Författare :Mary C Hunt; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Acyl-CoA thioesterase; acyl-CoA; peroxisome proliferator-activated receptor alpha; coenzyme A; bile acid metabolism; lipid metabolism.;

    Sammanfattning : The peroxisome proliferator-activated receptor alpha (PPARalpha) is a key nuclear receptor in the control of lipid metabolism. Target genes for PPARalpha include many enzymes involved in the oxidation of fatty acids in mitochondria and peroxisomes. LÄS MER

  5. 5. Acyl-CoA thioesterases : auxiliary enzymes in peroxisomal lipid metabolism

    Författare :Maria A K Westin; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Peroxisomes are small organelles essential for life, which carry out a variety of functions mostly related to lipid metabolism, including α- and β-oxidation as well as the first steps of plasmalogen biosynthesis. Peroxisomal β-oxidation metabolizes many different lipids including very long-chain fatty acids, dicarboxylic fatty acids, branched-chain fatty acids, eicosanoids and certain xenobiotic fatty acids.
