Sammanfattning : The overall aim of this thesis was to investigate the mechanisms beyond the interaction of cholinesterase inhibitors (ChEIs) with the cholinergic system in brain, with emphasis on their regulatory and mechanistic effects on subtypes of muscarinic and nicotinic receptors and neuroprotective properties. To investigate muscarinic receptor subtypes in frontal cortices of AD brains, the muscarinic antagonists pirenzepine, AF-DX 116, HHSiD, p-F-HHSiD and himbacine were used in receptor binding assays. Sammanfattning : Alzheimer’s disease (AD) is clinically characterised by an insidious onset with progressive symptoms of memory impairment, dysphasia, dyspraxia, dygnosia and visuospatial difficulties. 4. Early diagnosis of Alzheimer's disease : focus on quantitative EEG in relation to genetic, biochemical and neuroimaging markers
Sammanfattning : The main aims of this thesis were to explore whether quantitative EEG (qEEG) variables could be considered as positive markers of Alzheimer's disease (AD) and sensitive indices of disease progression and treatment effects, as well as to determine their relation to currently existing genetic, biochemical and neuroimaging markers. The main findings of seven subsequent studies are summarized as follows: 1) Combined alpha and theta relative power and temporo-parietal alpha band coherence were the best combination of qEEG variables for the discrimination of mild to moderate AD patients and healthy controls, giving 89% overall correct classification accuracy. 5. Functional features of human cytochrome P450 1A2 : with special focus on caffeine and melatonin metabolism
Sammanfattning : CYP1A2 is one of the major cytochrome P450 enzymes in the liver and metabolises drugs such as caffeine, clozapine, tacrine, and theophylline. CYP1A2 also has a role in chemical carcinogenesis due to its activation of procarcinogens, such as heterocyclic amines and arylamines. Caffeine is extensively used to assess CYP1A2 activity. LÄS MER
