    Författare :Daniel Decker; Leszek A. Kleczkowski; Henrik Vibe Schelle; Umeå universitet; []
    Nyckelord :LANTBRUKSVETENSKAPER; AGRICULTURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Chemical library screening; Cell wall synthesis; Glycosylation; Nucleotide sugars; Oligomerization; Protein structure; Reverse chemical genetics; Sugar activation; UDP-sugar synthesis; biokemisk farmakologi; biochemical pharmacology; biokemi; Biochemistry; biology; biologi; molekylärbiologi; Molecular Biology;

    Sammanfattning : UDP-sugar metabolizing pyrophosphorylases provide the primary mechanism for de novo synthesis of UDP-sugars, which can then be used for myriads of glycosyltranferase reactions, producing cell wall carbohydrates, sucrose, glycoproteins and glycolipids, as well as many other glycosylated compounds. The pyrophosphorylases can be divided into three families: UDP-Glc pyrophosphorylase (UGPase), UDP-sugar pyrophosphorylase (USPase) and UDP-N-acety lglucosamine pyrophosphorylase (UAGPase), which can be discriminated both by differences in accepted substrate range and amino acid sequences. 2. 2. Structural and Biophysical Studies of Nucleic Acids

    Författare :Wimal Pathmasiri; Adolf Gogoll; Lena Mäler; Uppsala universitet; []
    Nyckelord :Organic chemistry; nucleic acids; Nuclear Magnetic Resonance; molecular dynamics; sugar modified nucleosides; pKa; Organisk kemi;

    Sammanfattning : This thesis is based on six research publications concerned with (i) study of the molecular structures and dynamics of modified nucleosides; (ii) investigation of the effect of incorporation of modified nucleosides on the structure of DNA; (iii) examination of the effect of the sugar modifications on the pseudo-aromatic properties (pKa) of the nucleobases; (iv) analysis of the effect of the CH-π interactions on the relative stability of the DNA-RNA hybrid duplexes. The structural stability of the nucleic acids as well as their behavior in molecular recognition is dominated by hydrogen bonding and stacking interactions beside other non-covalent interactions. 3. 3. Macrocyclic Carbohydrate/Amino Acid Hybrid Molecules - Synthesis and Evaluation as Artificial Receptors

    Författare :Johan Billing; Centrum för analys och syntes; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; 3-dipolar cycloaddition; 3-triazoles; 2; 1; molecular recognition; artificial receptors; sugar amino acids; cyclic peptides; macrocycles; Organic chemistry; Organisk kemi;

    Sammanfattning : Methods were developed for the synthesis of three different types of macrocyclic carbohydrate/amino acid hybrid molecules. In the synthesis of the first type of macrocycles, a glucosamine derivative was oxidized at C6 and the obtained sugar amino acid was coupled to tripeptides. LÄS MER

  4. 4. Studies towards ß-turn mimetics, synthesis of kedarosamine from threonine, and synthesis of glycopeptides from mucins and gp120

    Författare :Tatjana Vuljanic; Centrum för analys och syntes; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Tn antigen; glycopeptides; glycosylation; diastereselective reduction; kedarosamine; kedarcidin chromophore; amino acids; peptide T; ß-turn mimetic; Organisk kemi; gp120; Organic chemistry;

    Sammanfattning : The secondary structure of proteins is crucial for proper function of the protein in biological systems. Peptide T, an octapeptide from the envelope glycoprotein gp120 of the HIV virus, has previously been shown to be responsible for binding to the CD4 receptor on T helper/inducer lymphocytes. LÄS MER

  5. 5. Steroids and bile acids in intrahepatic cholestasis of pregnancy : effects of treatment with ursodeoxycholic acid

    Författare :Ling-Jie Meng; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : A method permitting simultaneous analysis of individual steroid hormone metabolites and bile acids in different forms of conjugation in serum and urine was established. Total bile acids and neutral steroids were extracted from human serum or urine with octadecylsilane-bonded silica, separated into different groups of conjugates by anion-exchange chromatography and analyzed by fast atom bombardment mass spectrometry.
