  1. 1. The Tyrosine Kinase GTK : Signal Transduction and Biological Function

    Författare :Cecilia Annerén; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell biology; Protein tyrosine kinases; GTK; SHB; proliferation; survival; differentiation; β cells; pancreatectomy; cytokines; diabetes; PC12 cells; NGF; streptozotocin; focal adhesion kinase; RAP1; Cellbiologi; Cell biology; Cellbiologi; medicinsk cellbiologi; Medical Cell Biology;

    Sammanfattning : Protein tyrosine kinases play an important role in the regulation of various cellular processes such asgrowth, differentiation and survival. GTK, a novel SRC-like cytoplasmic tyrosine kinase, was recently cloned from a mouse insulinoma cell line and the present work was conducted in order to find a biological function of GTK in insulin producing and neuronal cells. 2. 2. Investigations of Strategies to Counteract Proinflammatory Cytokines in Experimental Type 1 Diabetes

    Författare :Andreas Börjesson; Stellan Sandler; Annika Carlsson; Jens Nielsen; Uppsala universitet; []
    Nyckelord :LANTBRUKSVETENSKAPER; AGRICULTURAL SCIENCES; Type 1 diabetes; proinflammatory cytokines; SOCS-3; pancreatic islets; inducible nitric oxide synthase; insulin secretion; NOD mice; islet transplantation; proinsulin conversion; streptozotocin; prolactin; Medicine; Medicin;

    Sammanfattning : Type 1 diabetes (T1D) is a chronic autoimmune disease targeted against the pancreatic β-cells. Proinflammatory cytokines are considered to play a major role in the destruction of the insulin-producing β-cells. This thesis studied strategies to counteract proinflammatory cytokines in experimental T1D. LÄS MER

  3. 3. MEKK-1 and NF-κB Signaling in Pancreatic Islet Cell Death

    Författare :Dariush Mokhtari; Nils Welsh; Michael Welsh; Anders Tengholm; Shanta Persaud; Uppsala universitet; []
    Nyckelord :Cell biology; MEKK-1; JNK; human islets; β-cells; siRNA; apoptosis; cell death; MAPK; nitric oxide; cytokines; streptozotocin; c-Rel; NF-κB; Cellbiologi;

    Sammanfattning : Type 1 diabetes is an autoimmune disease resulting in the selective destruction of the insulin producing β-cells in the pancreas. Pro-inflammatory cytokines and the free radical nitric oxide (NO) have been implicated in mediating the destruction of β-cells, possibly through activation of the mitogen activated protein kinases (MAPKs) JNK, ERK and p38. 4. 4. Role of Inducible Nitric Oxide Synthase and Melatonin in Regulation of β-cell Sensitivity to Cytokines

    Författare :Annika K. Andersson; Stellan Sandler; Nils Welsh; Décio L. Eizirik; Nils Billestrup; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell biology; β-cell; Cyclooxygenase; Cytokine; Diabetes; IFN-γ; IL-1β; iNOS; Insulin; Melatonin; Nitric oxide; Pancreatic islets; Phospholipase D; Prostaglandin E2; Streptozotocin; Cellbiologi; Cell biology; Cellbiologi;

    Sammanfattning : The mechanisms of β-cell destruction leading to type 1 diabetes are complex and not yet fully understood, but infiltration of the islets of Langerhans by autoreactive immune cells is believed to be important. Activated macrophages and T-cells may then secrete cytokines and free radicals, which could selectively damage the β-cells. LÄS MER

  5. 5. Experimental Studies Aiming to Prevent Type 1 Diabetes Mellitus

    Författare :Tobias Rydgren; Stellan Sandler; Nils Welsh; Adrian Bone; Uppsala universitet; []
    Nyckelord :Cell biology; Diabetes; Interleukin-1 trap; Simvastatin; Nitric oxide synthase; Cytokine; Pancreatic islet; Streptozotocin; Transplantation; Cellbiologi;

    Sammanfattning : Type 1 diabetes mellitus (T1DM) is an autoimmune disease in which T-cells and macrophages invade the islets of Langerhans and selectively destroy the insulin producing β-cells, either directly or through the secretion of e.g. cytokines and nitric oxide (NO). This thesis has studied possible strategies to prevent T1DM. LÄS MER
