6. DNA and RNA base analogue FRET - from fluorophore design to biochemical applications
Sammanfattning : This thesis focuses on the development and use of fluorescent base analogues (FBAs). They are important tools in research concerning nucleic acids structure, dynamics and interactions. LÄS MER
7. Functional characterization of the small antisense RNA MicA in Escherichia coli
Sammanfattning : The Escherichia coli small RNA (sRNA) MicA was identified recently in a genomewide search for sRNAs. It is encoded between the genes gshA and luxS in E. coli and its close relatives. The function of sRNAs in bacteria is generally believed to be in maintenance of homeostasis via stress-induced modulation of gene expression. LÄS MER
8. Conformationally Constrained Oligonucleotides for RNA Targeting
Sammanfattning : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. LÄS MER
9. On Transcriptome Sequencing
Sammanfattning : This thesis is about the use of massive DNA sequencing to investigate the transcriptome. During recent decades, several studies have made it clear that the transcriptome comprises a more complex set of biochemical machinery than was previously believed. LÄS MER
10. Ribosomal RNA Modification Enzymes : Structural and functional studies of two methyltransferases for 23S rRNA modification in Escherichia coli
Sammanfattning : Escherichia coli ribosomal RNA (rRNA) is post-transcriptionally modified by site-specific enzymes. The role of most modifications is not known and little is known about how these enzymes recognize their target substrates. LÄS MER
