  1. 1. Linear and Branched Chitosan Oligomers as Delivery Systems for pDNA and siRNA In Vitro and In Vivo

    Författare :Mohamed Mahmoud Issa; Per Artursson; Arto Urtti; Uppsala universitet; []
    Nyckelord :Medical sciences; Chitosan oligomers; gene delivery; gene expression; pDNA; siRNA; MEDICIN OCH VÅRD;

    Sammanfattning : In this thesis, chitosan, a biocompatible polysaccharide that has been approved as a food additive was selected as a platform for the development of safe, efficient non-viral gene delivery systems to mammalian cells. Previously, chitosan-based gene formulations had been generally associated with high molecular weight chitosans, which were poorly characterised in terms of molecular weight distribution and degree of acetylation. 2. 2. Chimeric gene delivery vectors : Design, synthesis, and mechanisms from transcriptomics analysis

    Författare :Moataz Dowaidar; Ülo Langel; Sandrine Sagan; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Cell-penetrating peptides; magnetic nanoparticles; graphene oxide; autophagy; siRNA; SCO; QSAR; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : Delivery of nucleic acid is a promising approach for genetic diseases/disorders. However, gene therapy using oligonucleotides (ONs) suffers from low transfection efficacy due to negative charges, weak cellular permeability, and enzymatic degradation. LÄS MER

  3. 3. Characterization of nucleic acid delivery with fatty acid modified cell-penetrating peptide nanoparticle formulations

    Författare :Tõnis Lehto; Ülo Langel; Patrick Midoux; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Transfection; gene delivery; oligonucleotide; complexes; non-viral; in vivo; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : Recent advances with techniques used for manipulating gene expression have brought us to an era where various gene therapeutic approaches are becoming common therapeutic tools for many previously incurable diseases. The main factor impeding the wider translation of gene therapy is that the active pharmaceutical ingredients used for interfering with gene expression are based on nucleic acids and synthetic oligonucleotides and such molecules do not readily reach their intracellular targets due to their physicochemical properties and therefore they require delivery vectors to cross the cell membrane. 4. 4. Regulating Gene Expression to Promote Osteoblastic Differentiation of Stem Cells

    Författare :Maruthibabu Paidikondala; Oommen P. Varghese; Jöns Hilborn; Laura Creemers; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Kemi med inriktning mot polymerkemi; Chemistry with specialization in Polymer Chemistry;

    Sammanfattning : Bone is a tissue that heals by itself, unless the defect is too large (critical size). Today, novel regenerative medicine approaches have emerged as an alternative to treat such defects. This thesis explores alternative therapeutic strategies for bone tissue engineering which are biocompatible and clinically translatable. LÄS MER

  5. 5. Nucleoside analoge cytotoxicity-focus on enzyme regulation, metabolism, and mechanisms of resistance

    Författare :Anna Fyrberg; Kourosh Lotfi; Curt Peterson; Freidoun Albertioni; Francoise Bontemps; Linköpings universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacology; Farmakologi;

    Sammanfattning : The aim of this thesis was to determine the role of nucleoside analog activating and deactivating enzymes in nucleoside analog metabolism and resistance development. Nucleoside analogs are anti-cancer drogs and are often used to treat different leukemias, attributably to presence of high levels of nucleoside analog activating enzymes in hematopoietic cells.
