    Sammanfattning : In microscale separations, such as capillary electrophoresis and -liquid chromatography, the liquid flow rates are in the order of nanoliters per second. If such flow rates are to be interfaced with a mass spectrometer (MS) using electrospray (ES) ionization, without loss of separation efficiency, each fraction of the analyte zone must remain undisturbed by the high voltage contact necessary for ES.     Sammanfattning : This thesis describes the current status and novel aspects of the analysis of the molecules of life, i.e. peptides and proteins, using capillary electrophoresis (CE) coupled to mass spectrometry (MS) via (sheathless) electrospray ionization (ESI). In this thesis, aspects on improved compatibility between liquid based separation systems and mass spectrometric detection were investigated regarding buffers, sample preparation and analysis of polar compounds as well as peptides and protein digests for enhanced ESI-MS performance.     Sammanfattning : Over the last decades the ongoing work in the fields of Lab-on-a-Chip and Micro-Total-Analysis-Systems has led to the discovery of new or improved ways to handle and analyse small volumes of biofluids and complex biosuspensions. The benefits of working on the microscale include: miniaturization of the analysis systems with less need for large sample volumes; temporal and spatial control of suspended particle/cell positions; low volume sheath flow lamination or mixing; novel separation techniques by using forces inherent to the microscale domain; precise regulation of sample temperatures and rapid analysis with less volumes needed to be processed.
