1. Genotyping and Mutation Detection In Situ : Development and application of single-molecule techniques
Sammanfattning : The human body is composed of trillions of cells closely working together to maintain a functional organism. Every cell is unique in molecular composition and can acquire genetic variations that might cause it to turn pathological. LÄS MER
2. Detection of immunoglobulin heavy chain gene rearrangement with PCR for MRD analysis in lymphoproliferative disorders
Sammanfattning : Immunoglobulin heavy chain (IGH) gene rearrangement occurs during early B-lymphocyte differentiation, assembling the different IGH gene segments to a functional gene, which can serve as a marker for study of lineage association and detection of Minimal Residual Disease (MRD) in clonal diseases deriving from B-lymphocytes or their early differentiation stages. Use of a molecular marker for the leukemic cells could help improve treatment by monitoring therapeutic efficacy, predicting relapse, and identifying very small amounts of tumour cells contaminating autografts after purging or enrichment of stem cells. 3. Ex‘PLA’ining the progression of pathological proteins in Alzheimer’s and Parkinson’s diseases : see(d)ing is believing
Sammanfattning : Alzheimer’s disease (AD) and Parkinson’s disease (PD) are the two most common forms of neurodegenerative disorders affecting approximately 50 million people worldwide. The underlying neuropathological processes leading to AD and PD share many similarities, i.e. aberrant protein aggregation of tau and alpha-synuclein (αSyn) in the brain. LÄS MER
4. Minimal Residual Disease Assessment in Childhood Acute Lymphoblastic Leukemia
Sammanfattning : Traditionally, response to treatment in hematological malignancies is evaluated by light microscopy of bone marrow (BM) smears, but due to more effective therapies more sensitive methods are needed. Today, detection of minimal residual disease (MRD) using immunological and molecular techniques can be 100 times more sensitive than morphology. LÄS MER
5. Proximity Ligation as a Universal Protein Detection Tool
Sammanfattning : Among the great challenges in biology are the precise quantification of specific sets of proteins and analyses of their patterns of interaction on a much larger scale than is possible today. This thesis presents a novel protein detection technique - proximity ligation - and reports the development and application of a nucleic acid amplification technique, RCA.
