21. Choice of tRNA on Translating Ribosomes
Sammanfattning : This thesis addresses different aspects of the question about accuracy of protein synthesis: i) the mechanism of tRNA selection during translation ii) study of ribosomal mutations that affect accuracy and iii) the choice of aminoacyl-tRNA isoacceptors on synonymous codons.By measuring the codon reading efficiencies of cognate and near-cognate ternary complexes we demonstrate that in optimal physiological conditions accuracy of substrate selection is much higher than previously reported; that during translation the ribosomal A site is not blocked by unspecific binding of the non-cognate tRNAs which would inhibit the speed of protein synthesis. 22. Adaptive Evolution of the Bacterial Translation Machinery
Sammanfattning : The process of protein synthesis via translation is of paramount importance for the existence of life on Earth. The bacterial translation machinery has embraced more than 3.5 billion years of molecular evolution to adapt and function efficiently under the provided physiological conditions. LÄS MER
23. From Structure to Function with Binding Free Energy Calculations for Codon Reading, Riboswitches and Lectins
Sammanfattning : Molecular association is part of many important processes in living cells. Computational methods for calculating binding free energies allows for a quantitative examination of biomolecular structures and hypotheses drawn from biochemical experiments. LÄS MER
24. Dynamics in Protein Synthesis, Structural Studies of Translation Factors
Sammanfattning : Protein biosynthesis is performed on ribosomes. The ribosome is a complex of ribosomal RNA and proteins forming two subunits. A messenger RNA strand is decoded on the small subunit by charged aminoacyl transfer RNA (aa-tRNA) molecules and each tRNA molecule delivers its amino acid to the growing polypeptide chain on the large subunit. 25. Ribosomal translocation in real time : Method development to Applications
Sammanfattning : Translational elongation is the process in which the ribosome adds one amino acid at a time to the nascent peptide chain. As the ribosome elongates the peptide chain by 14 - 20 amino acids per second and performs hundreds of such cycles per protein, ‘elongation’ is one of the most crucial steps in translation. LÄS MER
