16. Induction of apoptosis in relation to chromatin structure and inhibition of replication by DNA damage from ionizing radiation
Sammanfattning : The theme of this thesis has been chromatin organization ranging from methodological studies to involvement in apoptotic response. The aims have been: (i) to compare the information obtained by the AVTD method and comet assay concerning DNA-loop organization, (ii) to test the hypothesis that DNA-loop relaxation could be the triggering signal for induction of apoptosis in G0-lymphocytes, and (iii) to study the dose response for inhibition of the replication fork movement and pathways for the DNA repair process at the replication fork. Sammanfattning : Mitochondria are cytoplasmic organelles in eukaryotic cells, mainly devoted to the synthesis of ATP, through the Oxydative Phospshorylation System (OXPHOS). The organelle has its own genome, the mitochondrial DNA (mtDNA) that encodes for all the RNA components of the mitochondrial translation apparatus (two rRNAs and 22 tRNAs) and for 13 subunits of the OXPHOS complexes. Sammanfattning : Mitochondria are a dynamic network of subcellular organelles that produce the majority of cellular ATP through the process of oxidative phosphorylation (OXPHOS). The components of the respiratory chain are encoded by two separate genomes, nuclear DNA and mitochondrial DNA (mtDNA), and the proper maintenance of both of these genomic entities is therefore crucial for cellular ATP levels and the survival of the cell.
