  1. 1. Replication Fork Stability in Mammalian Cells

    Författare :Ingegerd Elvers; Thomas Helleday; Jan H. J. Hoeijmakers; Stockholms universitet; []
    Nyckelord :replication fork progression; replication fork stability; re-priming; DNA damage signalling; NATURAL SCIENCES; NATURVETENSKAP; molekylärgenetik; Molecular Genetics;

    Sammanfattning : Maintaining replication fork integrity is vital to preserve genomic stability and avoid cancer. Physical DNA damage and altered nucleotide or protein pools represent replication obstacles, generating replicative stress. Numerous cellular responses have evolved to ensure faithful DNA replication despite such challenges. LÄS MER

  2. 2. Induction of apoptosis in relation to chromatin structure and inhibition of replication by DNA damage from ionizing radiation

    Författare :Jesper Torudd; Mats Harms-Ringdahl; Mats Ljungman; Stockholms universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ionizing radiation; DNA-loop; replication fork; Toxicology; Toxikologi;

    Sammanfattning : The theme of this thesis has been chromatin organization ranging from methodological studies to involvement in apoptotic response. The aims have been: (i) to compare the information obtained by the AVTD method and comet assay concerning DNA-loop organization, (ii) to test the hypothesis that DNA-loop relaxation could be the triggering signal for induction of apoptosis in G0-lymphocytes, and (iii) to study the dose response for inhibition of the replication fork movement and pathways for the DNA repair process at the replication fork. 3. 3. Insights into the roles of the essential Pfh1 DNA helicase in the nuclear and mitochondrial genomes

    Författare :Parham Pourbozorgi Langroudi; Nasim Sabouri; Anders Hofer; Per Sunnerhagen; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; G4; genome integrity; Pfh1; Top1; helicase; topoisomerase; replication; transcription;

    Sammanfattning : Eukaryotic cells have two sets of genomes, the nuclear and mitochondrial, and both need to be accurately maintained. Also, the rate of transcription must be precisely regulated in these genomes. However, there are many natural barriers that dysregulate these processes. LÄS MER

  4. 4. Homologous recombination at replication forks in mammalian cells

    Författare :Cecilia Lundin; Thomas Helleday; Klaus Erixon; Keith W. Caldecott; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Genetics; Genetik;

    Sammanfattning : Accumulating evidence indicates that homologous recombination plays an important role in mammalian cells, primarily in the reactivation of stalled replication forks. During a normal round of replication, stalling of the replication fork occurs frequently when a nick or a lesion, which cannot be by-passed, is encountered. LÄS MER

  5. 5. Linking transcription to DNA replication in mammalian mitochondria

    Författare :Yonghong Shi; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Most of the adenosine phosphate (ATP) used to drive cellular processes is produced through oxidative phosphorylation (OXPHOS). At the heart of this process is the electron transport chain, which contains a number of protein complexes located in the inner membrane of mitochondria. LÄS MER
