  1. 1. Protein engineering to explore and improve affinity ligands

    Författare :Martin Linhult; KTH; []
    Nyckelord :binding; affinity; human serum albumin HSA ; albumin-binding domain ABD ; affinity chromatography; deamidation; protein A; stabilization; Z-domain; capacity; protein G; cleaning-in-place CIP ; protein engineering; C2 receptor;

    Sammanfattning : In order to produce predictable and robust systems forprotein purification and detection, well characterized, small,folded domains descending from bacterial receptors have beenused. These bacterial receptors, staphylococcal protein A (SPA)and streptococcal protein G (SPG), possess high affinity to IgGand / or HSA. LÄS MER

  2. 2. Deciphering Binding Patterns of Therapeutic Antibodies with Immune Cells : From Method Development to Application

    Författare :Sina Bondza; Jos Buijs; Marika Nestor; Mark Cragg; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; affinity; binding kinetics; CD20; cell-based assay; immunology; receptor-ligand interaction; therapeutic antibody; Immunologi; Immunology; Medical Cell Biology; Medicinsk cellbiologi;

    Sammanfattning : Reversible binding, for example between signaling molecules and receptors on the cell surface, is one of the main means to communicate information in cellular systems. Knowledge about how molecules interact is crucial for both understanding biological function and for therapeutic intervention. LÄS MER

  3. 3. Studies of the Neuropeptide Y Receptor Y2 in Human and Zebrafish

    Författare :Helena Fällmar; Dan Larhammar; Ingrid Lundell; Nina Mohell; Raquel E. Rodríguez Rodríguez; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Y2-receptor; NPY; PYY; G-protein-coupled receptor; mutagenesis; receptor binding; zebrafish; evolution; Pharmacology; Farmakologi; Farmakologi; Pharmacology;

    Sammanfattning : The G-protein coupled receptors (GPCRs) comprise the largest family of receptors in humans and other vertebrates. They are embedded in the cell membrane and are activated by many different signaling molecules. Activation modulates cellular signal transduction pathways and influences many physiological processes. 4. 4. Binding Kinetics of Proteins at Immune-Cell Contacts

    Författare :Manto Chouliara; NanoLund: Centre for Nanoscience; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; T cells; Binding Kinetics; Affinity; Lifetime; CD4; CD2; TIRF Microscopy; SLBs;

    Sammanfattning : Protein-protein interactions are crucial in numerous cellular functions and biological processes that take place inside our body. It is therefore not surprising that these interactions also govern the response of our body´s defence mechanism, the so-called immune system, towards an infection. LÄS MER

  5. 5. Computational Modeling of the AT2 Receptor and AT2 Receptor Ligands : Investigating Ligand Binding, Structure–Activity Relationships, and Receptor-Bound Models

    Författare :Christian Sköld; Anders Karlén; Anders Hallberg; Torbjörn Lundstedt; Tommy Liljefors; Uppsala universitet; []
    Nyckelord :Pharmaceutical chemistry; Angiotensin II; AT1; AT2; SAR; bioactive conformation; turn mimetic; peptidomimetic; DISCO; homology model; 3D-QSAR; CoMFA; Farmaceutisk kemi;

    Sammanfattning : Rational conversion of biologically active peptides to nonpeptide compounds with retained activity is an appealing approach in drug development. One important objective of the work presented in this thesis was to use computational modeling to aid in such a conversion of the peptide angiotensin II (Ang II, Asp-Arg-Val-Tyr-Ile-His-Pro-Phe).
