1. Xenograft Rejection : A Study of Cytokine mRNA Expression in Experimental Models
Sammanfattning : Acute cellular xenograft rejection of ICCs triggered mRNA expression of proinflammatory cytokines (IL-1b and TNF-a), Th1 associated cytokines (IL-12p40, IL-2 and IFN-g) and Th2 associated cytokines (IL-4 and IL-10). The peak values and the kinetics of the mRNA expression indicated that acute cellular xenograft rejection is enhanced by the proinflammatory response and mediated by the Th1 associated response, whereas the function of the Th2 associated response most likely is to counteract the Th1 associated cytokines and thereby inhibit bystander damage by the delayed type hypersensitivity like rejection process. 4. HAMLET - In vivo effects and mechanisms of tumor-cell death
Sammanfattning : HAMLET (human alpha-lactalbumin made lethal to tumor cells), a molecular complex derived from human milk, is an interesting new tool in cancer research since it induces programmed cell death in tumor cells while leaving normal, differentiated cells unharmed. 5. Islet xenograft rejection : studies in the pig-to-rodents and pig-to-primate models
Sammanfattning : The aims of this thesis was: 1) to compare islet allo- and xenograft rejection in rats, using immunosuppression with CsA in a mixed allogeneic-xenogeneic islet transplantation model, 2) to evaluate the efficacy of various immunosuppressive drugs, with special reference to Leflunomide, in preventing islet xenograft rejection in the pig-to-rat model, 3) to perform an immunohistochemical study of porcine islet xenograft rejection in primates including primates treated with CsA+DSG, and 4) to study the metabolism and excretion of porcine C-peptide in mice, with special reference to the use of donor-specific C-peptide for monitoring islet xenograft function.
