  1. 1. EMBRYONIC GLIA AND FOREBRAIN NEUROGENESIS; in vitro generation and differentiation of telencephalic neurons

    Författare :Charlotta Skogh; Neurobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; beta tubulin III; GFAP; RC2; Neurology; neuropsychology; neurofysiologi; DARPP-32; retinoic acid; serum; EGF; co-culture; cell culture; striatum; LGE; progenitor; precursor; radial glia; region specific; development; CNS; mouse; human; neurophysiology; Neurologi; neuropsykologi;

    Sammanfattning : The current thesis is aimed to increase our understanding of the generation and differentiation of telencephalic neurons, with emphasis on the role of glial cells in these processes. Using in vitro methods we have studied the role of glial cells in neurogenesis, inheritance of genetic regional specification, and influence on differentiation of neurons. LÄS MER

  2. 2. Characterization of human dopaminergic neurons in the developing mesencephalon and upon differentiation of stem cells- for replacement therapy in Parkinson´s disease

    Författare :Jenny Nelander Wahlestedt; Utvecklings- och regenerativ neurobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Parkinson´s disease; stem cells; ventral mesencephalon; dopaminergic neurons; cell replacement therapy; human fetal tissue; radial glia; microRNA; BAC recombineering; reporter cell lines;

    Sammanfattning : Cell-replacement therapy is a promising approach for treating patients with Parkinson ́s disease (PD). For this purpose, there is a need for developing a protocol that can generate high numbers of human transplantable mesencephalic dopaminer- gic (mesDA) neurons. LÄS MER

  3. 3. On the influence of glia on neurite outgrowth from dopamine neurons in the nigrostriatal system

    Författare :Malin Saga Johansson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Dopamine; glia; astrocytes; olfactory ensheathing cells; development; ni-grostriatal; mesencephalon; Parkinson s disease; 6-hydroxydopamine;

    Sammanfattning : Interactions between glial cells and neurons are important during fetal development, in the maintenance of the normal functions in tile central nervous system (CNS), arid in response to injuries or diseases. Some of die motor disturbances in Parkinson's disease, a neurodegenerative disorder where one of the hallmarks is degeneration of dopamine (DA) neurons and concomitant depletion of DA in striatum, can be alleviated by intrastriatal grafting of embryonic DA tissue. 4. 4. Nuclear receptor and Wnt function in developing dopaminergic neurons

    Författare :Kyle Matthew Sousa; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The progressive degeneration of dopaminergic neurons in the substantia nigra is one of the major pathological events associated with Parkinson’s Disease. Dopaminergic (DA) cell replacement therapy has emerged in recent years as a possible approach towards treating Parkinsonism. LÄS MER

  5. 5. Wnt signalling in the development of ventral midbrain dopaminergic neurons

    Författare :Gonçalo de Sá e Sousa de Castelo-Branco; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Dopaminergic Neuron; Ventral Midbrain Development; Writs; Dickkopf; beta-catenin; GSK-3beta; Stem Cells; Neural Precursors;

    Sammanfattning : Dopaminergic (DA) neurons in the ventral midbrain (VM) are one of the major cell types lost in Parkinson's disease (PD). Proof-of-principle exists for cell replacement therapies for PD, but wider application is halted by the unavailability of abundant sources of DA neurons. Stem cells might constitute one of these sources. LÄS MER
