  1. 1. Pulmonary Drug Absorption : In vitro and in vivo investigations of drug absorption across the lung barrier and its relation to drug physicochemical properties

    Författare :Ann Tronde; Gerrit Borchard; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; lung; pulmonary; drug delivery; inhalation; drug transport; pharmacokinetics; absorption; bioavailability; drug metabolism; preclinical; rat; in vivo; isolated and perfused lung; Caco-2; nebulization catheter; aerosol; physicochemical; peptide; efflux; FARMACI; PHARMACY; FARMACI; Biopharmaceutics; biofarmaci;

    Sammanfattning : Although, pulmonary drug delivery is a well established means for targeting of drugs to the lungs for the treatment of respiratory diseases as well as for the systemic delivery of volatile anesthetic agents, drug absorption kinetics in the lung have not been subjected to extensive research. The main objective of this thesis was to investigate drug absorption characteristics of the lung barrier, using the isolated and perfused rat lung model and in vivo pharmacokinetic studies in rats. 2. 2. Lipid-based liquid crystals as drug delivery vehicles for antimicrobial peptides

    Författare :Lukas Boge; Martin Andersson; Lovisa Ringstad; RISE; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Liquid crystals; liquid crystalline nanoparticles; cubosome; hexosome; phase behavior; antimicrobial peptide; AMP; glycerol monooleate; proteolysis; antimicrobial effect; antimicrobial resistance; bacterial membrane; membrane interaction; infection; topical delivery; pulmonary delivery;

    Sammanfattning : The development of antimicrobial resistance is a great challenge within health sectors worldwide. Thus, demand for new, efficient treatments is urgent in order to treat various bacterial infections. Antimicrobial peptides (AMPs) are a group of antibiotics that have gained more and more attraction in the past decade. LÄS MER

  3. 3. Synchronized delivery of inspired nitric oxide : Effects on oxygenation and pulmonary tension during artificial ventilation

    Författare :Erkki Heinonen; Tim Higenbottam; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell biology; nitric oxide; pulsed administration; pulmonary hypertension; oxygenation; equine; Cellbiologi; Cell biology; Cellbiologi; Physiology; fysiologi;

    Sammanfattning : Nitric oxide (NO) is a mediator of vascular smooth muscle tension that metabolises rapidly in blood. NO delivered by inhalation can therefore be used as a selective pulmonary vasodilator to relieve pulmonary hypertension or to improve oxygenation with no systemic effects. LÄS MER

  4. 4. Transport and metabolism of opioid tetrapeptide agonists with a focus on oral and respiratory delivery

    Författare :Eva Krondahl; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; opioid peptides; permeability; metabolism; oral delivery; pulmonary delivery; FARMACI; PHARMACY; FARMACI; galenisk farmaci; Pharmaceutics;

    Sammanfattning : The clinical development of peptide drugs for oral delivery has often been hindered because of poor transport across the intestinal membranes and degradation in the gastrointestinal tract and liver. Alternative delivery routes through more permeable membranes preferably with less pronounced proteolytic activity, such as the nasal and pulmonary routes, have therefore been considered. LÄS MER

  5. 5. Characterisation of an in vitro dissolution method for assessment of novel pulmonary drug delivery systems : With a focus on controlled release systems

    Författare :Irès van der Zwaan; Göran Frenning; Per Hansson; Rebecca Fransson; Alessandra Rossi; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pulmonary drug delivery; in vitro dissolution; mechanistic model; controlled release; physiologically based biopharmaceutics modelling;

    Sammanfattning : Pulmonary drug delivery has been used for decades to treat local diseases like asthma. When using the pulmonary route to deliver drugs, several important lung features are being used, such as a large surface area available for absorption, high organ vascularization, and a thin blood-alveolar barrier.
