    Författare :Axel Klaesson; Ola Söderberg; Mats Nilsson; Cristina Al-Khalili Szigyarto; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; In situ proximity ligation assay PLA ; rolling circle amplification RCA ; Hybridization chain reaction HCR ; proxHCR; Oligonucleotide design; Protein-protein interactions; Post-translational modifications.; Farmaceutisk vetenskap; Pharmaceutical Science; Molekylär cellbiologi; Molecular Cellbiology;

    Sammanfattning : In situ proximity ligation assay (PLA) is a method for detection of protein interactions, post-translational modifications (PTMs) and individual proteins that allows information about their localization in a cell or tissue to be extracted. The method is based on oligonucleotide-conjugated antibodies (proximity probes) that upon binding of two epitopes in close proximity give rise to an amplifiable DNA circle. LÄS MER

  2. 22. Identification of SLiMs: Mapping and characterizing motif-based protein interactions

    Författare :Muhammad Ali; Ylva Ivarsson; Shoshana Wodak; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Phage display; protein-protein interactions; short linear motifs; PDZ; FERM; affinity; x-ray crystallization; molecular docking; NLS;

    Sammanfattning : During the last twenty years it has become evident that about 35-40% of amino acids in the proteome are in regions that have evolved to remain unstructured. These intrinsically disordered regions contain short linear motifs (SLiMs), which serve as docking sites for protein-protein interactions. LÄS MER

  3. 23. Evolution and viral mimicry of short linear motif-mediated interactions

    Författare :Filip Mihalič; Per Jemth; Leo James; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; phage display; protein-protein interactions; large-scale discovery; short linear motifs; viral mimcry; viral hijacking; Biokemi; Biochemistry; Medical Biochemistry; Medicinsk biokemi;

    Sammanfattning : Proteins are one of the most fundamental building blocks of life and their interactions regulate every cellular process. Historically they have been conceptualized as predominantly folded entities with well-defined secondary and tertiary structures. LÄS MER

  4. 24. The solution conformations of macrocycles. Applications in the exploration of weak interactions and in drug development

    Författare :Emma Danelius; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Macrocycles; cyclic peptides; NMR; solution conformational analysis; NAMFIS; β-hairpin; weak interactions; halogen bonding; protein-protein interaction; bioactive conformation; macrolides; cell permeable conformation.;

    Sammanfattning : Understanding the solution conformation and dynamics of molecules with biological relevance, as well as the impact of their conformation stabilizing weak interactions, is for example important for drug design. Macrocycles have attractive pharmaceutical properties, and are of special interest as drug leads for targets with large, flat and featureless binding sites like protein-protein interfaces. LÄS MER

  5. 25. Fluorescence Studies of Membranes -- Proteins and Lipids, their Dynamics and Interactions

    Författare :Johan Strömqvist; Jerker Widengren; Thorsten Wohland; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Fluorescence correlation spectroscopy; cytokinesis; collision theory; protein-protein interactions; lipid dynamics; single-molecule experiment; Biological physics; Biologisk fysik;

    Sammanfattning : In this thesis, fluorescence spectroscopy was utilized to probe protein and lipid dynamics and interactions in their native, or close to native, environments. Thereby insight could be gained into the fundamentals of bacterial cell division and the innateimmune system. A particular focus has been devoted to fluorescence fluctuations. LÄS MER
