  1. 1. Structure determination and thermodynamic stabilization of an engineered protein-protein complex

    Författare :Elisabet Wahlberg; Per-Åke Nygren; Bengt-Harald Jonsson; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; affibody; protein structure; coupled folding; NMR spectroscopy; protein stability; protein-protein interactions; binding thermodynamics; isothermal titration calorimetry; protein engineering; Structural biochemistry; Strukturbiokemi;

    Sammanfattning : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain. 2. 2. Perplexing Protein Puzzles

    Författare :Stina Lindman; Biofysikalisk kemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; calbindin D9k; split-GFP; protein G B1; pKa-values; protein reconstitution; Electrostatic interactions; protein stability; protein denaturation; heteronuclear NMR; pH titrations;

    Sammanfattning : Protein structure and stability are inherent in the amino acid sequence and governed by non-covalent interactions. The cooperation between forces is, however, perplexing and not well understood. In order to elucidate and predict protein folding and stability, detailed studies of non-covalent interactions are required. LÄS MER

  3. 3. Unravelling protein stability and aggregation

    Författare :Tanja Weiffert; NanoLund: Centre for Nanoscience; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; protein stability; protein aggregation; amyloid beta peptide; protein stabilization; split GFP; monellin; secretagogin; ; aggregation kinetics; toxicity; aggregation mechanism; coaggregation;

    Sammanfattning : Proteins are complex macromolecules that are fundamental to all living species. The stability of proteins are governed by non-covalent forces within the protein and between the protein and its surrounding environment. LÄS MER

  4. 4. SOD1 Aggregation : Relevance of thermodynamic stability

    Författare :Lisa Lang; Mikael Oliveberg; Michele Vendruscolo; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Aggregation; misfolding; Superoxide dismutase SOD1 ; Amyotrophic lateral sclerosis ALS ; unfolding; thermodynamic stability; Biochemistry; biokemi;

    Sammanfattning : Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease affecting the upper and lower motor neurons causing muscle atrophy and paralysis followed by death. Aggregates containing superoxide dismutase (SOD1) are found as pathological hallmark in diseased ALS patients. LÄS MER

  5. 5. Protein stability and mobility in live cells : Revelation of the intracellular diffusive interaction organization mechanisms

    Författare :Xin Mu; Mikael Oliveberg; Johan Elf; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; diffusive interactions; macromolecular crowding; protein thermodynamic stability; protein mobility; in-cell NMR; Hofmeister series; Biochemistry; biokemi;

    Sammanfattning : Biochemical processes inside living cells take place in a confined and highly crowded environment. As such, macromolecular crowding, one of the most important physicochemical properties of cytoplasm, is an essential element of cell physiology. LÄS MER
