  1. 6. Design and Synthesis of Aspartic and Serine Protease Inhibitors : Targeting the BACE-1 and the HCV NS3 Protease

    Författare :Fredrik Wångsell; Mats Larhed; Jari Yli-Kauhaluoma; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer s disease; BACE-1; transition state mimetic; tertiary hydroxyl group; hydroxyethylene; statine; hydroxyethylamine; hepatitis C; HCV NS3; bioisostere; protease inhibitor.; Pharmaceutical chemistry; Läkemedelskemi; Medicinal Chemistry; Läkemedelskemi;

    Sammanfattning : This thesis describes work done to design and synthesize protease inhibitors, with the intention of developing therapeutic agents for Alzheimer’s disease (AD) and the chronic liver condition caused by infection of the hepatitis C virus (HCV). AD is the most common form of dementia, and HCV infection is the primary reason for liver transplantation in industrialized countries. LÄS MER

  2. 7. Characterization of conjugated protease inhibitors

    Författare :Erika Billinger; Gunnar Johansson; Boris Turk; Uppsala universitet; []
    Nyckelord :Serine protease inhibitors; conjugation; immobilization; leupeptin analogues; potato serine protease inhibitor; soluble carriers; inorganic carriers.; Biokemi; Biochemistry;

    Sammanfattning : The overall theme of this thesis is a step by step approach for the design and characterization of conjugated protease inhibitors. This involves both a new assay method for protease activity and protease inhibition (paper I), a study of the stoichiometry for protease inhibitor interaction (paper II), design of inhibitory peptides (paper IV) and the construction of inhibitor conjugates (paper III & IV). 3. 8. Structure-aided design of antiviral drugs : Application of the method on HIV-1 protease and SIV reverse transcriptase

    Författare :Hans Ola Andersson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Developmental biology; HIV; protease; SIV; reverse transcriptase; structure; inhibitor; drug design; Utvecklingsbiologi; Developmental biology; Utvecklingsbiologi; molekylärbiologi; Molecular Biology;

    Sammanfattning : Many efforts have been made to control the AIDS epidemic. Extensive studies have been done on the biology, biochemistry, and structural biology of HIV in the search for antiviral drugs. The viral-encoded enzymes reverse transcriptase and protease have been main targets for drug design. LÄS MER

  4. 9. Tertiary Alcohol- or β-Hydroxy γ-Lactam-Based HIV-1 Protease Inhibitors : Microwave Applications in Batch and Continuous Flow Organic Synthesis

    Författare :Per Öhrngren; Mats Larhed; Knud J. Jensen; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; HIV; AIDS; protease inhibitor; aspartic protease; lactam; tertiary alcohol; continuous-flow; microwave; non-resonant; MAOS; Medicinal Chemistry; Läkemedelskemi;

    Sammanfattning : Since the outbreak of the HIV/AIDS pandemic in the 1980s, the disease has cost the lives of over 30 million people, and a further 33 million are currently living with the HIV infection. With the appropriate treatment, HIV/AIDS can today be regarded as a chronic but manageable disease. LÄS MER

  5. 10. Development of the gastrointestinal tract in young mammals : Effects of enteral provocation with protease or phytohaemagglutinin in neonatal rats

    Författare :Ester Arevalo Sureda; Funktionell zoologi; []
    Nyckelord :gut; Intestine; Pancreas; development; precocious; protease; PHA; enterocytes; permeability; endocytosis; IgG; FcRn; Blimp1; T-lymphocyte; passive immunity; altricial; neonatal; suckling; athymic; rat; immunohistochemistry IHC ;

    Sammanfattning : The rat, as an altritial species, is born with an immature gastrointestinal tract and intestinal barrier function, which is highly absorptive to milk-borne bioactive molecules that can pass undigested and reach the general circulation of the suckling newborn. This passage occurs by the neonatal-Fc-receptor (FcRn) binding and trancytosis of immunoglobulin G in the proximal small intestine (SI) and by the highly endocytic vacuolated enterocytes non-selectively in the distal SI.
