  1. 1. Antibody based plasma protein profiling

    Författare :Ulrika Qundos; Jochen M. Schwenk; Peter Nilsson; Mathias Uhlén; Ulrike Korf; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; protein profiling; plasma; antibody; affinity proteomics; biomark- ers; multiplex; assay development;

    Sammanfattning : This thesis is about protein profiling in serum and plasma using antibody suspension bead arrays for the analysis of biobanked samples and in the context of prostate cancer biomarker discovery. The influence of sample preparation methods on antibody based protein profiles were investigated (Papers I-III) and a prostate cancer candidate biomarker identified and verified (Papers III-V). Sammanfattning : Affinity-based proteomics utilizes affinity binders to detect target proteins in a large-scale manner. This thesis describes a high-throughput method, which enables the search for biomarker candidates in human plasma and serum. LÄS MER

  4. 4. Multivariate profiling of metabolites in human disease : Method evaluation and application to prostate cancer

    Författare :Elin Thysell; Henrik Antti; Janne Lehtiö; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; metabolite profiling; metabolomics; predictive metabolomics; mass spectrometry; GC-MS; biomarkers; chemometrics; design of experiments; multivariate data analysis; prostate cancer; bone metastases; plasma; biologisk kemi; biological chemistry;

    Sammanfattning : There is an ever increasing need of new technologies for identification of molecular markers for early diagnosis of fatal diseases to allow efficient treatment. In addition, there is great value in finding patterns of metabolites, proteins or genes altered in relation to specific disease conditions to gain a deeper understanding of the underlying mechanisms of disease development. 5. 5. Multiplexed absolute quantification of blood plasma proteins using targeted proteomics

    Författare :David Kotol; Mathias Uhlén; Edfors Fredrik; Roman Zubarev; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; mass spectrometry; targeted proteomics; stable isotope standards; recombinant protein standards; absolute quantification; blood plasma; multiplex analysis; plasma profiling; Biotechnology; Bioteknologi;

    Sammanfattning : Proteins are the molecular building blocks of all living organisms. They are the functioning actors in metabolism and communication, and they give specific architecture and purpose to all tissues. Their abundance is dynamic and can be interpreted and associated to different physiological states or diseases. LÄS MER
