    Författare :Johan Kjellström; Bröstcancer-genetik; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Inorganic chemistry; polyelectrolyte; medium effects; salt dependence; DSC; CD; HPLC; oligonucleotide; hairpin; DNA; anticancer active platinum II compounds; kinetics; mechanism; Oorganisk kemi; Cytology; oncology; cancerology; Cytologi; onkologi; cancer;

    Sammanfattning : The kinetics and mechanism for interactions of the anticancer active platinum(II) complexes cis-[PtCl2(NH3)2)], cis-[PtCl2(NH3)(c-C6H11NH2)], and trans-[PtCl2-(NH3)(quinoline)] with DNA have been studied using a combination of HPLC, circular dichroism (CD), UV/vis spectroscopy, and differential scanning calorimetry (DSC). Reactions of the monoaqua forms (charge = +1) of these complexes with single-stranded DNA oligonucleotides, typically between 13-17 bases long, were found to depend on i) the composition of the reaction medium, ii) the exact nature and location of the platination target, and iii) on the DNA structure. 3. 8. Biosensor technology applied to hybridization analysis and mutation detection

    Författare :Peter Nilsson; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; biosensor; genosensor; surface plasmon resonance; hybridization; nucleic acids; oligonucleotide; mutation detection; mismatch discrimination; modular; Bioengineering; Bioteknik;

    Sammanfattning : This thesis demonstrates the application of biosensor technology for molecular biology investigations, utilizing a surface plasmon resonance based optical device for mass sensitive detection of biomolecular interactions at a chipsurface. Oligonucleotide model systems were designed for analysis of the action of DNA manipulating enzymes. LÄS MER

  4. 9. Quantitative NanoSIMS provides subcellular concentration and distribution of oligonucleotide therapeutics

    Författare :Cécile Becquart; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NanoSIMS; Antisense Oligonucleotide; Intracellular concentration;

    Sammanfattning : Antisense oligonucleotides (ASOs) represent a powerful therapeutic modality that can selectively modulate gene expression. However, ASOs face two major hurdles that restrict their use in the clinic. The first issue is delivery of the ASO to a tissue of therapeutic interest while reducing exposure to unrelated tissues. 5. 10. Characterization of nucleic acid delivery with fatty acid modified cell-penetrating peptide nanoparticle formulations

    Författare :Tõnis Lehto; Ülo Langel; Patrick Midoux; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Transfection; gene delivery; oligonucleotide; complexes; non-viral; in vivo; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : Recent advances with techniques used for manipulating gene expression have brought us to an era where various gene therapeutic approaches are becoming common therapeutic tools for many previously incurable diseases. The main factor impeding the wider translation of gene therapy is that the active pharmaceutical ingredients used for interfering with gene expression are based on nucleic acids and synthetic oligonucleotides and such molecules do not readily reach their intracellular targets due to their physicochemical properties and therefore they require delivery vectors to cross the cell membrane.
