  1. 1. Olfactory Transfer of Analgesic Drugs After Nasal Administration

    Författare :Ulrika Espefält Westin; Erik Björk; Eva Brittebo; Margareta Hammarlund-Udenaes; Paolo Colombo; Uppsala universitet; []
    Nyckelord :Pharmaceutics; Nasal administration; Olfactory transfer; Olfactory pathways; Central nervous system; Blood-brain barrier; Nasal respiratory mucosa; Olfactory mucosa; Olfactory bulb; Horizontal Ussing chamber; Morphine; Dihydroergotamine; Rat; Mouse; Swine; Autoradiography; Viability; Powder formulations; Galenisk farmaci;

    Sammanfattning : Nasal administration of analgesics for achieving rapid pain relief is currently a topic of great interest. The blood-brain barrier (BBB) restricts access to the central nervous system (CNS) for several central-acting drugs, such as morphine and dihydroergotamine, which results in a substantial effect delay. 2. 2. Tissue-Selective Activation and Toxicity of Substituted Dichlorobenzenes : Studies on the Mechanism of Cell Death in the Olfactory Mucosa

    Författare :Anna Franzén; Eva Brittebo; Ingvar Brandt; Olavi Pelkonen; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Toxicology; tissue-selective toxicity; bioactivation; olfactory mucosa; substituted dichlorobenzenes; Bowman s glands; sustentacular cells; ER stress; Grp78; caspase-12; caspase-3; CYP2A5; protein adduction; nasal toxicity; Toxikologi; Toxicology; Toxikologi;

    Sammanfattning : The nasal passages are constantly exposed to both air- and bloodborne foreign compounds. In particular, the olfactory mucosa is demonstrated to be susceptible to a variety of drugs and chemicals. LÄS MER

  3. 3. Nasal administration of compounds active in the central nervous system : Exploring the olfactory system

    Författare :Maria Dahlin; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; Nasal administration; olfactory pathway; cerebrospinal fluid; autoradiography; FARMACI; PHARMACY; FARMACI; farmaceutisk farmakologi; Pharmaceutical Pharmacology;

    Sammanfattning : The nasal administration of drugs offers advantages over administration by intravenous injection. Drugs can be rapidly absorbed through the nasal mucosa, resulting in a rapid onset of action, and also avoiding degradation in the gastrointestinal tract and first-pass metabolism in the liver. 4. 4. Neurotoxic Effects of Dichlorophenyl Methylsulphones Related to Olfactory Mucosal Lesions

    Författare :Carina Carlsson; Ingvar Brandt; Hans Tjälve; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biology; olfcatory toxicity; olfactory bulb; aryl methylsulphone; neurotoxicity; QSAR; Biologi; Biology; Biologi; Ecotoxicology; Ekotoxikologi;

    Sammanfattning : This thesis deals with the highly potent olfactory mucosa toxicant 2,6-dichlorophenyl methylsulphone (2,6-diClPh-MeSO2) and its non-toxic 2,5-chlorinated isomer (2,5-diClPh-MeSO2). In mice, both substances bind firmly in the olfactory mucosa and the olfactory bulb, which are important components of the sensory system. LÄS MER

  5. 5. Models for the Transfer of Drugs from the Nasal Cavity to the Central Nervous System

    Författare :Björn Jansson; Erik Björk; Sveinbjörn Gizurarson; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; Administration; intranasal; Nasal mucosa; Central nervous system; Olfactory bulb; Rat; Mouse; Swine; Cattle; Diffusion chambers; culture; In vitro; Biological models; Biological transport; Dextrans; Dopamine; Gels; Polysaccharides; Autoradiography; Fluorescence microscopy; Biofarmaci; Biopharmacy; Biofarmaci;

    Sammanfattning : The blood-brain barrier restricts the access of many compounds, including therapeutic agents, to the brain. Several human studies indicate that nasal administration of hydrophilic compounds, such as peptides, can bypass the blood-brain barrier. The aims of this thesis were to develop and refine models for this direct nose-to-brain transfer. LÄS MER
