13. Regulation of chromatin transitions in the 3D architecture of the nucleus
Sammanfattning : The thesis explores the dynamic features and functions of chromatin crosstalk in the 3D nucleus space, with a focus on the regulation of chromatin transitions. In Paper I we employed the circular chromosome conformation capture assay (4C) and Chromatin in situ Proximity (ChrISP) technique to uncover an inter-chromosomal chromatin fiber interactome comprising both transcriptionally active circadian genes and repressed lamina-associated domains (LADs). 14. Crosstalk between environmental signals and 3d genome organization in the regulation of gene expression
Sammanfattning : The thesis explores the connection between environmental stimuli and gene expression regulated by the spatial changes in genome organization. In Paper I, by applying state of the art Circular Chromosome Conformation Capture assay (4C) and Chromatin in situ Proximity (ChrISP) techniques, we show that transcriptionally active circadian genes meet in space with repressed lamina-associated domains (LADs), and that these interactions are under the control of the circadian clock. 15. Dynamics of higher order chromatin structures
Sammanfattning : During the last few decades, the intensive focus on microscopy observations and genome sequencing analyses has proved that the genomic DNA is packaged in the non-random manner in the nucleus of interphase cells. Accumulated evidence have thus documented that the chromatin organization in 3D plays key roles in central biological processes, such as transcription, replication and DNA repair. LÄS MER
