  1. 1. Benefits of Non-Linear Mixed Effect Modeling and Optimal Design : Pre-Clinical and Clinical Study Applications

    Författare :Charles Ernest II; Andrew Hooker; Mats Karlsson; Bart Ploeger; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacometrics; optimal design; nonlinear mixed effects models; population models; Pharmaceutical Science; Farmaceutisk vetenskap;

    Sammanfattning : Despite the growing promise of pharmaceutical research, inferior experimentation or interpretation of data can inhibit breakthrough molecules from finding their way out of research institutions and reaching patients. This thesis provides evidence that better characterization of pre-clinical and clinical data can be accomplished using non-linear mixed effect modeling (NLMEM) and more effective experiments can be conducted using optimal design (OD). 2. 2. Effect Oriented Planning in Military Mission Support Systems : Models and Heuristic Approaches

    Författare :Kristian Lundberg; Kaj Holmberg; Torbjörn Larsson; Stefan Engevall; Linköpings universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : Today there are many aspects of model based planning, probably much more than for just 20 years ago. Today data is not the problem, data is everywhere, but the big issue is to understand how to gain advantage of data in decision making - a growing focus is now on modeling!This work is devoted to this task and can be seen as a three part study. 3. 3. Nonlinear Mixed Effects Modeling of Combination Therapies in Oncology

    Författare :Marcus Baaz; Stiftelsen Fraunhofer-Chalmers Centrum för Industrimatematik (FCC); []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacol- ogy; Radiation Therapy; Oncology; Mathematical Modeling; Combination Therapy; Nonlinear Mixed Effects;

    Sammanfattning : It has been shown that simultaneous administration of two or more anticancer agents can have a beneficial effect on cancer patients. This type of treatment is called combination therapy and is nowadays commonly used in the fight against cancer. LÄS MER

  4. 4. Pharmacometric Models to Improve Treatment of Tuberculosis

    Författare :Elin M Svensson; Mats O Karlsson; Maria C Kjellsson; Ulrika S H Simonsson; N. L’Ntshotsholé (Shasha) Jumbe; Uppsala universitet; []
    Nyckelord :pharmacokinetics; pharmacodynamics; population approach; nonlinear mixed-effects models; multidrug-resistant tuberculosis; bedaquiline; antiretroviral; drug-drug interactions; time-to-event; albumin; Klinisk farmakologi; Clinical Pharmacology;

    Sammanfattning : Tuberculosis (TB) is the world’s most deadly infectious disease and causes enormous public health problems. The comorbidity with HIV and the rise of multidrug-resistant TB strains impede successful therapy through drug-drug interactions and the lack of efficient second-line treatments. LÄS MER

  5. 5. Methods for handling model misspecification in pharmacometric model-based approaches for the analysis of longitudinal clinical trial data

    Författare :Estelle Chasseloup; Mats Karlsson; Andrew Hooker; Karl Brendel; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacometrics; nonlinear mixed-effects models; misspecification; type I error; power; accuracy; placebo; individual model averaging; IMA; likelihood ratio test; LRT; model averaging; covariate selection; mixture models; visual predictive checks; VPC; avatars; digital twins; virtual individual; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : Pharmacometrics model-based analyses proved to be increasingly useful in drug discovery and development. This usefulness was made evident by the formalisation of the model-integrated drug discovery and development framework (MID3). LÄS MER
