1. Chitosan Polyplexes as Non-Viral Gene Delivery Systems : Structure-Property Relationships and In Vivo Efficiency
Sammanfattning : The subject of this thesis was to develop and optimize delivery systems for plasmid DNA (pDNA) based on biocompatible polymers, in particular chitosan, suitable for non-viral gene therapy. At the onset of this thesis, studies had reported conflicting results on the efficiency of chitosan-based gene delivery systems. LÄS MER
2. Characterization of nucleic acid delivery with fatty acid modified cell-penetrating peptide nanoparticle formulations
Sammanfattning : Recent advances with techniques used for manipulating gene expression have brought us to an era where various gene therapeutic approaches are becoming common therapeutic tools for many previously incurable diseases. The main factor impeding the wider translation of gene therapy is that the active pharmaceutical ingredients used for interfering with gene expression are based on nucleic acids and synthetic oligonucleotides and such molecules do not readily reach their intracellular targets due to their physicochemical properties and therefore they require delivery vectors to cross the cell membrane. 3. Dendrimer-mediated gene delivery: Effect of polymer structure on cellular response
Sammanfattning : The delivery of nucleic acids to cells, in vitro and in vivo, is becoming a realistic approach for intervention with gene expression at a molecular level. It can be used to repair malfunctioning genes or to introduce new functionalities, with the most important application being clinical treatment of human diseases. LÄS MER
4. Enhancing the efficiency of viral and non-viral gene delivery vectors
Sammanfattning : Gene therapy, which involves the introduction, alteration, or deletion of genetic material within cells to treat or prevent disease, holds great promise for addressing various genetic disorders and other medical conditions. However, several challenges remain in the field, particularly concerning nucleic acid delivery into target cells. LÄS MER
5. Endocytosis of therapeutic macromolecules in tumor cells - Mechanistic aspects of the proteoglycan receptor function
Sammanfattning : Novel therapeutics to combat cancer are urgently needed. Most current pharmacological therapies have severe side effects and are seldom curative. Macromolecular drugs, and in particular nucleic acid based drugs, offer a potential remedy for this situation. LÄS MER
