1. Reactivity of human omental blood vessels. Effects of 5-hydroxytryptamine, substance P and the intravenous anaesthetic propofol
Sammanfattning : Regulation of blood flow involves several endogenous substances and mechanisms, and exogenous substances may also affect the blood flow. In the present thesis, human omental blood vessels were investigated in vitro concerning (i) characteristics of 5-hydroxytryptamine (5-HT) receptors; (ii) mediators of substance P (SP)-induced relaxation; (iii) direct and indirect (via effects on sympathetic neurotransmission) effects of the intravenous anaesthetic propofol. 2. Vasodilator actions of EDHF and anandamide
Sammanfattning : Vasodilator responses to endothelium-derived hyperpolarizing factor (EDHF) and anandamide in isolated arteries were investigated. A combination of the potassium channel blockers charybdotoxin and apamin inhibits EDHF-mediated relaxations. LÄS MER
3. Histamine receptors and neuropeptides in the cranial circulation
Sammanfattning : The sensory neuropeptides substance P (SP) and calcitonin gene-related peptide (CGRP) are present in human cerebral, meningeal and temporal arteries, as are peptides associated with the sympathetic (neuropeptide Y, NPY) and parasympathetic (vasoactive intestinal peptide, VIP) nervous system. NPY is a vasoconstrictor in cerebral and meningeal arteries and potentiates noradrenaline (NA) as a vasoconstrictor in temporal arteries. LÄS MER
4. PACAP and orphanin FQ/nociceptin -Distribution, importance and regulation in sensory neurons and spinal cord
Sammanfattning : Neuropeptide expression in the nervous system is abundant and plastic, and an altered expression after injury is an example of a functional response, helping the neuron to cope with adverse changes, and involves effects on differentiation, synthesis, repair/regeneration, survival, and modulation of signal transmission. I have investigated the distribution/expression of the neuropeptides, orphanin FQ/nociceptin (OFQ/N) and pituitary adenylate cyclase activating polypeptide (PACAP), under normal conditions, in response to nerve injuries, and also studied sensory responses in mice deficient for the PAC1 receptor (PAC1-/-). 5. PACAP and airway regulation with special reference to smooth muscle and neutrophils
Sammanfattning : The investigations focus on the neuropeptidepituitary adenylate cyclase-activating peptide and its effects on airway regulation, with emphasis on smooth muscle relaxation and neutrophil activation and migration. In the nasal mucosa, PACAP-containing nerve fibers were localized invascular smooth muscle and close to seromucous glands. LÄS MER
