1. Molecular principles of protein stability and protein-protein interactions
Sammanfattning : Proteins with highly specific binding properties constitute the basis for many important applications in biotechnology and medicine. Immunoglobulins have so far been the obvious choice but recent advances in protein engineering have provided several novel constructs that indeed challenge antibodies. LÄS MER
2. Structural Transitions in Helical Peptides : The Influence of Water – Implications for Molecular Recognition and Protein Folding
Sammanfattning : Fluctuations in protein structure are vital to function. This contrasts the dominating structure-function paradigm, which connects the well-defined three-dimensional protein structure to its function. However, catalysis is observed in disordered enzymes, which lack a defined structure. LÄS MER
3. Conformational changes in Staphylococcus aureus' alpha-toxin : from water-soluble monomer to a transmembrane channel
Sammanfattning : Alpha-toxin is secreted by Staphylococcus aureus as a soluble monomer that without further activation can oligomerize and form a transmembrane channel. One of the aims of the present work was to understand the underlying mechanisms of the membrane insertion process. LÄS MER
4. Structure determination and thermodynamic stabilization of an engineered protein-protein complex
Sammanfattning : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain. 5. Structural and thermodynamical basis for molecular recognition between engineered binding proteins
Sammanfattning : The structural determination of interacting proteins, both as individual proteins and in their complex, complemented by thermodynamical studies are vital in order to gain in-depth insights of the phenomena leading to the highly selective protein-protein interactions characteristic of numerous life processes. This thesis describes an investigation of the structural and thermodynamical basis for molecular recognition in two different protein-protein complexes, formed between so-called affibody proteins and their respective targets.
