  1. 1. Contributions to nonlinear mixed-effects modeling

    Författare :Jacob Leander; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; nonlinear mixed-effects models; stochastic differential equations; parameter estimation; extended Kalman filter; sensitivity equations; stochastic differential equations;

    Sammanfattning : The topic of this thesis is nonlinear mixed-effects models. A nonlinear mixed-effects model is a hierarchical regression model used to analyze measurements from several individuals simultaneously, which allows sharing of information between similar individuals. LÄS MER

    Sammanfattning : It has been shown that simultaneous administration of two or more anticancer agents can have a beneficial effect on cancer patients. This type of treatment is called combination therapy and is nowadays commonly used in the fight against cancer. LÄS MER

  3. 3. Mixed Effects Modeling of Deterministic and Stochastic Dynamical Systems - Methods and Applications in Drug Development

    Författare :Jacob Leander; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; pharmacometrics; pharmacokinetics; mixed effects; drug development; pharmacodynamics; parameter estimation; time-to-event; mathematical modeling; dynamical systems;

    Sammanfattning : Mathematical models based on ordinary differential equations (ODEs) are commonly used for describing the evolution of a system over time. In drug development, pharmacokinetic (PK) and pharmacodynamic (PD) models are used to characterize the exposure and effect of drugs. LÄS MER

  4. 4. Nonlinear Mixed Effects Methods for Improved Estimation of Receptor Occupancy in PET Studies

    Författare :Matts Kågedal; Mats Karlsson; Andrew C Hooker; Gitte Knudsen; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; PET; positron emission tomography; receptor occupancy; nonlinear mixed effects; NONMEM; optimal design; dose finding; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Sammanfattning : Receptor occupancy assessed by Positron Emission Tomography (PET) can provide important translational information to help bridge information from one drug to another or from animal to man. The aim of this thesis was to develop nonlinear mixed effects methods for estimation of the relationship between drug exposure and receptor occupancy for the two mGluR5 antagonists AZD9272 and AZD2066 and for the 5HT1B receptor antagonist AZD3783. 5. 5. Development and Evaluation of Nonparametric Mixed Effects Models

    Författare :Paul Baverel; Mats Karlsson; Radojka Savic; Nils Hoem; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; nonparametric; model; pharmacometrics; pharmacokinetics; pharmacodynamic; imprecision; covariate analysis; parameter distribution; Statistics; computer and systems science; Statistik; data- och systemvetenskap; Pharmaceutical pharmacology; Farmaceutisk farmakologi; Biopharmaceutics; Biofarmaci; Clinical Pharmacology; Klinisk farmakologi; Computer Systems; Datorteknik; Farmakologi; Pharmacology; Statistics; Statistik;

    Sammanfattning : A nonparametric population approach is now accessible to a more comprehensive network of modelers given its recent implementation into the popular NONMEM application, previously limited in scope by standard parametric approaches for the analysis of pharmacokinetic and pharmacodynamic data. The aim of this thesis was to assess the relative merits and downsides of nonparametric models in a nonlinear mixed effects framework in comparison with a set of parametric models developed in NONMEM based on real datasets and when applied to simple experimental settings, and to develop new diagnostic tools adapted to nonparametric models.
