1. Minor groove binding unsymmetrical cyanine dyes : binding and dissociation studies in free solution and in electrophoresis gels

2. Detection of Biomolecules: Syntheses and Studies of Minor Groove Binding Unsymmetrical Cyanine Dyes and Chromogenic Enzyme Substrates
Sammanfattning : Visualisation or detection of biomolecules is usually achieved by the use of fluorescent or highly coloured chromophores. The work presented in this thesis deals with the development of new chromogenic substrates for the detection of enzyme activity, and new DNA-binding fluorescent dyes for staining of DNA. LÄS MER
3. Biophysical studies of DNA binding – by the large filament-forming protein Rad51 and the small minor-groove binder Hoechst 33258
Sammanfattning : Mechanistic insight into the nature of DNA-binding ligands is crucial for both drug development as well as understanding more complex biological reactions that take place in the cell. In this Thesis, two rather different DNA-binding molecules are considered: 1. LÄS MER
4. NMR structral studies on ruthenium complexes binding to DNA
Sammanfattning : The Ru-complex Λ,Λ-[bidppz(bipy)4Ru2]4+ (Λ,Λ-B) exhibits multiple modes of interactions with the palindromic DNA dodecamer [d(CGCGAATTCGCG)]2, however excluding any type of intercalation as shown by both optical spectroscopy and NMR. NMR observation of Λ,Λ-B titration into a DNA solution results in both a transient binding, with observation of fast exchange for protons located near the ends of the DNA duplex, and a more stable interaction in the DNA minor groove indicated by a slow exchange mechanism and binding sites for G2, G4 and A5 of the same DNA strand. 5. Molecular Recognition Mechanisms: Thermodynamics and Polarization Spectroscopy of DNA--Intercalator Complexes
Sammanfattning : Mechanisms for molecular recognition in DNA--intercalator complexes have been investigated by optical spectroscopy. The effect of electrical charge on binding geometry and binding thermodynamics was studied using 2,7-diazapyrene (DAP) and its N-methylated cations DAP+ and DAP2+ as model ligands. LÄS MER
