1. Hepatic Disposition of Drugs and the Utility of Mechanistic Modelling and Simulation
Sammanfattning : The elimination of drugs from the body is in many cases performed by the liver. Much could be gained if an accurate prediction of this process could be made early in the development of new drugs. However, for the elimination to occur, the drug molecule needs first to get inside the liver cell. LÄS MER
2. Mechanistic Based Pharmacokinetic-Pharmacodynamics models for Drug Interactions and Disease Population Predictions
Sammanfattning : Therapeutic dose of a medication refers to the quantity of a drug required to produce a pharmacological effect without causing unacceptable adverse events. Dose selection in the clinical setting is not straight forward due to various factors, including specific patient factors such as age, sex, weight, genetic variants and renal/hepatic function, as well as external factors such as food and co-medication, all of which can influence the efficacy and safety of a drug. 3. Mechanistic-Empirical Modelling of Flexible Pavement Performance : Verifications Using APT Measurements
Sammanfattning : Mechanistic-Empirical (M-E) pavement design procedures are composed of a reliable response model to estimate the state of stress in the pavement and distress models in order to predict the different types of pavement distresses due to the prevailing traffic and environmental conditions. One of the main objectives of this study was to develop a response model based on multilayer elastic theory (MLET) with improved computational performance by optimizing the time consuming parts of the MLET processes. 4. Semi-mechanistic models of glucose homeostasis and disease progression in type 2 diabetes
Sammanfattning : Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by consistently high blood glucose, resulting from a combination of insulin resistance and reduced capacity of β-cells to secret insulin. While the exact causes of T2DM is yet unknown, obesity is known to be a major risk factor as well as co-morbidity for T2DM. LÄS MER
5. Characterization of the Mechanisms of Drug Release from Polymer-Coated Formulations using Experiments and Modelling
Sammanfattning : The main objectives of the work presented in this thesis were to develop new experimental methods to improve the understanding of the release mechanism from a coated formulation and to develop new mechanistic models to describe the release process for pellets coated with a semi-permeable film. Another aim was to characterize the release mechanism from a formulation coated with films made of a blend of a water-insoluble polymer, ethyl cellulose (EC), and a water-soluble polymer, hydroxypropyl cellulose (HPC).
