1. HIV and SIV specific cellular immunity in macaque models
Sammanfattning : Cellular immunity is believed to be an important prerequisite for an effective HIV vaccine. Accruing data from individuals able to contain HIV-1 replication during natural infection underscores the contribution of CD8+ T cells in viral control. LÄS MER
2. Hormonal regulation of the primate uterus : effects of hormone treatment and lifestyle intervention
Sammanfattning : The uterus is a major hormone-responsive reproductive organ in primates. Hormones promote their effect via different pathways. Receptors involved in steroid hormone, thyroid hormone and insulin signaling have been found in the endometrium. LÄS MER
3. Dissection of HIV-1 Env-specific B cell responses in nonhuman primates
Sammanfattning : Vaccine-induced protection is generally mediated by long-lived antigen-specific B cell responses. Most licensed vaccines target pathogens that display relatively low variability, but for highly variable pathogens, such as HIV-1, vaccine development is more challenging. LÄS MER
4. Repertoire diversity and maturation of HIV-1 vaccine-induced B cell responses
Sammanfattning : In the work presented in this thesis, I characterized B cell responses in rhesus macaques inoculated with HIV-1 envelope glycoprotein (Env)-based vaccines to define genetic and functional properties of the elicited antibodies for an improved understanding of Env immunogenicity. In Paper I, we investigated antibody heavy chain V gene segment expression in the overall B cell repertoire and in response to vaccination by using 2 approaches, 454-pyrosequencing and single-cell RT-PCR of bulk and sorted memory B cells, respectively. 5. Discovery of targets for tumor immunotherapy using phage display technology
Sammanfattning : This thesis has focused on the development of methods and their applications for the identification of antibodies reactive with tumor associated antigens (TAA). The phage display technology provides a powerful mean to generate antibodies potentially useful for tumor immunotherapy. LÄS MER
