    Sammanfattning : The atypical antipsychotic drug (APD) clozapine is the most efficacious APD in treatment-resistant schizophrenia including negative symptoms and cognitive impairment, and still lacks extrapyramidal side effects (EPS). Clozapine, which also possesses an antidepressant effect and can be used as monotherapy in bipolar disorder, has a broad receptor binding profile with higher affinity for the α2-adrenoceptor and several serotonergic receptors than D2 receptor, enhances dopamine output in the medial prefrontal cortex (mPFC) and facilitates glutamatergic NMDA receptor-mediated transmission in pyramidal cells in the same brain region. Generally, the so-called atypical APDs, such as clozapine, may in contrast to typical APDs improve not only positive, but also negative symptoms and some aspects of cognitive impairment associated with schizophrenia.     Sammanfattning : Non-competitive NMDA receptor antagonists, such as phencyclidine (PCP) and dizocilpine (MK-801) can even acutely induce a drug induced psychotic state which closely resembles schizophrenia. The midbrain dopamine (DA) neurons, which originate in the ventral tegmental area (VTA), are involved in motivational, attentional and cognitive processes, and have also been profoundly implicated in schizophrenia.     Sammanfattning : Introduction: In this thesis, MEGA-edited Magnetic Resonance Spectroscopy (MRS) has been used for the purpose of non-invasive detection of !- aminobutyric acid (GABA) within the brain.
