1. Protein Interaction Studies with Low Molecular Weight Ligands : Applications for Drug Discovery, Basic Research and Diagnostic Tool Design
Sammanfattning : In this thesis, the interactions between different proteins and small ligands were characterized by surface plasmon resonance spectroscopy (SPR) and fluorescence resonance energy transfer (FRET) based assays. For the C-reactive protein (CRP), a new type of artificial binder was identified which allows designing diagnostic assays superior to commonly used standard assays. LÄS MER
2. Ligand binding and enzyme catalysis studied by molecular dynamics simulations
Sammanfattning : Molecular dynamics simulations and free energy calculations can be applied to biomolecular systems to predict ligand binding affinities. Combined with hybrid quantum-classical potential energy surfaces, such computations can also be used to probe enzymatic catalysis mechanisms. LÄS MER
3. Studies of PDGF receptor signaling in vitro and in vivo
Sammanfattning : Platelet-derived growth factor receptor (PDGFR) signaling is essential for proliferation, migration and survival of cells of mesenchymal origin; however, its deregulation has been associated with various diseases, including cancer. The aim of this thesis was to clarify the molecular mechanisms of PDGFR signaling regulation. LÄS MER
4. Design and synthesis of HIV-1 protease inhibitors
Sammanfattning : Human Immunodeficiency Virus (HIV) is the causative agent of Acquired Immune Deficiency Syndrome (AIDS). The C2-symmetric HIV-1 protease is one of the prime targets for chemotherapy in the treatment of the HIV infection. Inhibition of HIV-1 protease leads to immature and non-infectious viral particles. 5. Experimental studies on thrombosis and thrombolysis : With special reference to importance of lys-plasminogen, active site thrombin inhibitors and stable fish oil
Sammanfattning : Dissolution and prevention of thrombus in the atherosclerotic coronary artery have become an important part of treatment therapy of acute myocardial infarction. However, the currently most effective thrombolytic agent, recombinant tissue-type plasminogen activator (rt-PA), only elicits 80% recanalization of thrombotically occluded coronary arteries and early occlusion occurs in about 40% of successfullyrecanalized vessels.
