  1. 1. Development of low affinity techniques for applications in clinical chemistry

    Författare :Lisa Leickt; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; chromatography performance; acute myocardial infarction; antibodies; creatine kinase MB isoforms; lectins; low affinity interactions; weak affinity chromatography; Clinical chemistry; Klinisk kemi;

    Sammanfattning : An increase in the plasma level of creatine kinase MB (CKMB) has long been used to indicate acute myocardial infarction (AMI). For optimal treatment an early diagnosis is important. An increase in the ratio between the two isoforms of CKMB is seen much earlier than an increase in the total level of CKMB. LÄS MER

  2. 2. Identification of SLiMs: Mapping and characterizing motif-based protein interactions

    Författare :Muhammad Ali; Ylva Ivarsson; Shoshana Wodak; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Phage display; protein-protein interactions; short linear motifs; PDZ; FERM; affinity; x-ray crystallization; molecular docking; NLS;

    Sammanfattning : During the last twenty years it has become evident that about 35-40% of amino acids in the proteome are in regions that have evolved to remain unstructured. These intrinsically disordered regions contain short linear motifs (SLiMs), which serve as docking sites for protein-protein interactions. LÄS MER

  3. 3. Development of Electroacoustic Sensors for Biomolecular Interaction Analysis

    Författare :Henrik Anderson; Ilia Katardjiev; Björn Ingemarsson; Inger Vikholm-Lundin; Uppsala universitet; []

    Sammanfattning : Biomolecular interaction analysis to determine the kinetics and affinity between interacting partners is important for the fundamental understanding of biology, as well as for the development of new pharmaceutical substances. A quartz crystal microbalance instrument suitable for kinetics and affinity analyses of interaction events was developed. LÄS MER

  4. 4. Affinity-, Partition- and Permeability Properties of the Human Red Blood Cell Membrane and Biomembrane Models, with Emphasis on the GLUT1 Glucose Transporter

    Författare :Christine Lagerquist Hägglund; Per Lundahl; Jørgen Vinten; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Affinity; Aromatic amino acids; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dihydrocytochalasin B; Dissociation constant; Drug absorption; Ethanol; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Specific; Streptavidin; Tyrosine; Tryptophan; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : The human glucose transporter GLUT1 is abundant in red blood cells, the blood-brain barrier and epithelial cells, where it mediates the transport of the energy metabolite, glucose. In the present work some properties of GLUT1, including affinity binding of both substrates and inhibitors, transport rates as well as permeabilities of aromatic amino acids and drug-membrane interactions were analyzed by chromatographic methods. 5. 5. Protein Interactions: Electrostatics and Ligand Binding

    Författare :Ingemar André; Biofysikalisk kemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Molecular biophysics; Molekylär biofysik; secondary structure formation; protein GB1; pKa values; pH stability; NMR; electrostatic interactions; M proteins; calbindin D9k; calcium binding; cooperativity; calmodulin; calbindin D28k;

    Sammanfattning : This thesis deals with Ca2+ binding to proteins, electrostatic interactions in and between proteins as well as inter- and intramolecular interactions. A computer program was developed to determine Ca2+ binding constants from experimental titration data of proteins. LÄS MER
